dc.description.abstractBu tez çalışmasında, Salvia chionantha Boiss. bitkisinin uçucu yağı GC ve GC/MS ile analiz edildi. Uçucu yağda 55 bileşen belirlendi ve 54 bileşenin yapısı aydınlatıldı. Germakren D (% 25.03), ß-karyofilen (% 8.71), Spathulenol (% 5.86) ve ?-humulen (% 4.82) ana bileşikler olarak tespit edildi. Uçucu yağ, hekzan, diklormetan, etil asetat ve metanol özütlerinin antioksidan aktiviteleri ? -karoten-linoleik asit ve DPPH serbest radikal giderim yöntemleri kullanılarak belirlendi. Etil asetat özütü ? -karoten-linoleik asit yönteminde, yüksek lipit peroksidasyon inhibisyon aktivitesi (% 87,4±1.01) gösterdi. DPPH yönteminde, özütler arasında etil asetat özütünün % 90,12±0.51 inhibisyon ile en iyi radikal giderim aktivitesine sahip olduğu tespit edildi. Ayrıca özütlerin toplam fenolik ve toplam flavonoid miktarları sırasıyla pirokatekol ve kersetine eşdeğer olarak belirlendi. S. chionantha'nın uçucu yağ ve özütlerinin in vitro antikolinesteraz aktivitesi Ellman metodu kullanılarak spektrofotometrik olarak saptandı. Uçucu yağda orta seviyede asetil- ve bütiril- kolinesteraz inhibitor aktivitesi (500 µg/ mL konsantrasyonda sırasıyla, %56,65±1.92 ve %41,68±2.91) görüldü. Özütler AChE enzimini inhibe edemedikleri halde BChE enzimini inhibe edebildikleri belirlendi.
dc.description.abstractIn this thesis, the essential oil of Salvia chionantha Boiss. was analyzed by GC and GC-MS. Totally, 55 components were detected in the essential oil and 54 components were elucidated. Germacrene D (25.03%), ß-caryophyllene (8.71 %), Spathulenol (5.86 %) and ?-humulene (4.82 %) were identified as the major compounds. The antioxidant activity of the essential oil, hexane, dichloromethane, ethyl acetate and methanol extracts was determined by using two complementary tests systems; namely, ? -carotene-linoleic acid and DPPH free radical scavenging assays. In ? -carotene-linoleic acid assay, ethyl acetate extract (% 87.4±1.01) showed great lipid peroxidation inhibition. In DPPH assay, among the extracts, the ethyl acetate extract (%90.12±0.51) showed better activity. In addition, total phenolic and total flavanoid contents of the extracts were determined as pyrocatechol and quercetin equivalents, respectively. The essential oil and the various extracts of S. chionantha were also assigned spectrofotometrically for their in vitro anticholinesterase activity by using Ellman method. The essential oil showed modarate acetyl- and butyryl-cholinesterase inhibitory activity (56.65±1.92% and 41.68±2.91% at 500 µg/mL concentration, respectively). However, the extracts were exhibited no activity against acetylcholinesterase enzyme, they only showed weak activity against butyrylcholinesterase enzyme.en_US
