RhoA/Rho-kinaz (ROCK) gen polimorfizmlerinin ve ROCK1 ve ROCK2 gen ekspresyonlarının Behçet hastalığı ile ilişkisinin araştırılması

Abstract

Bu çalışmanın amacı Türk toplumunda RhoA, Rho-kinaz (ROCK)1 ve ROCK2 gen polimorfizmlerinin ve bunun yanında ROCK1 ve ROCK2 gen ekspresyonlarının Behçet hastalığı (BH) ile ilişki olup olmadığını araştırmaktı. Çalışmaya 194 Behçet hastası ve benzer yaş ve cinsiyetteki 276 sağlıklı kontrol dahil edildi. Polimorfizmler genomik DNA'da, BioMark 96.96 Dynamic Array Sistem kullanılarak analiz edildi. ROCK1 ve ROCK2 gen ekspresyonu için kan örneklerinden mRNA ekstraksiyonu yapıldıktan sonra real time polimeraz zincir reaksiyonu uygulandı. RhoA geni rs6784820'in AG genotipi, A aleli, rs974495'in GA genotipi, rs2177268'in TA genotipi, rs3448'in T aleli BH olanlar ile kontrol grubu arasında anlamlı olarak farklı idi. ROCK1 geni rs2271255'in TC genotipi, C aleli, rs73963110'un TC, CC genotipi, C aleli, rs112130712'nin TC genotipi, C aleli, rs111874856'nın CT genotipi, T aleli, rs112108028 (Pro1164Leu)'in GA genotipi, A aleli, ROCK2 geni rs35768389 (Asp601Val)'un TA, AA genotipi, A aleli, rs1515219'un CT, TT genotipi, T aleli BH olanlar ile kontrol grubu arasında anlamlı derecede farklıydı (p<0.05). Bu polimorfizmler ile BH'nin klinik belirtileri arasında da anlamlı ilişki vardı. ROCK1 geni rs35996865, rs111312709 (Thr792Ala), ROCK 2 geni rs726843, rs2290156, rs965665, rs10178332, rs2230774, rs2230774 (Thr431Asn), rs2230774 (Thr431Ser), rs6755196, rs10929732, rs34945852 (Lys1083Met) ve RhoA geni rs2878298 polimorfizmleri alel ve genotip dağılımları BH olanlar ile kontrol grubu arasında anlamlı farklılık göstermedi. BH olanlarda ROCK2 mRNA ekspresyonunun kontrol grubuna göre anlamlı derecede yüksek olduğu görüldü (p<0.05). RhoA, ROCK1 ve ROCK2 gen polimorfizmlerinin BH ile ilişkisi ilk olarak ortaya konmuştur. Bu polimorfizmler Türk popülasyonunda BH'ye bireysel yatkınlığı belirleyebilir. Bu çalışmayla BH için yeni bir gen bölgesi belirlenmiştir.

The aim of this study is to investigate the association between RhoA, Rho-kinase (ROCK)1 and ROCK2 gene polymorphisms and also ROCK1, ROCK2 gene expressions and patients with Behçet?s disease (BD) in a Turkish population. A total of 194 BD patients and 276 healthy controls with similar age and sex were included to this study. Polymorphisms were analyzed in genomic DNA using a BioMark 96.96 dynamic array system. mRNA from blood samples was extracted, and real time polymerase chain reaction was performed for ROCK1 and ROCK2 gene expressions. AG genotype, A allele of rs6784820, GA genotype of rs974495, TA genotype of rs2177268, T allele of rs3448 for RhoA gene, TC genotype, C allele of rs2271255, TC, CC genotype, C allele of rs73963110, TC genotype, C allele of rs112130712, CT genotype, T allele of rs111874856, GA genotype, A allele of rs112108028 (Pro1164Leu) for ROCK1 gene, TA, AA genotypes, A allele of rs35768389 (Asp601Val), CT, TT genotypes, T allele of rs1515219 for ROCK2 gene were significantly different between patients and controls (p<0.05). There were significant associations between the genotype and allele frequencies of these polymorphisms and manifestations of BD. No marked associations were found with ROCK1 gene rs35996865, rs111312709 (Thr792Ala), ROCK 2 gene rs726843, rs2290156, rs965665, rs10178332, rs2230774, rs2230774 (Thr431Asn), rs2230774 (Thr431Ser), rs6755196, rs10929732, rs34945852 (Lys1083Met) and RhoA gene rs2878298 polymorphisms. There was a marked increase in peripheral blood mRNA ROCK2 expressions in patients (p<0.05). The association between BD and RhoA, ROCK1, ROCK2 gene polymorphism is showed firstly in this study. These polymorphisms may modify individual susceptibility to BD in the Turkish population. A new gene region with this study has been found to explain the pathıgenesis of BD.
