dc.description.abstractBu çalışmanın amacı depresyon hastalarında sertralinin farmakogenetik yönden araştırılması ile plazma ilaç ve metabolitinin tayinini gerçekleştirmektir. Bu sebeple sertralin farmakokinetiğini ve farmakodinamiğini etkileyen genlerdeki polimorfizmler sertralin tedavisi gören 64 hastada araştırılmıştır. Bu polimorfizmler, polimeraz zincir reaksiyonu ve restriksiyon parça uzunluk polimorfizmleri yöntemleriyle belirlenmiştir. Sertralin ve ana metaboliti desmetil sertralinin plazma konsantrasyonları yüksek performanslı svı kromatografisi ile ölçülmüştür.
dc.description.abstractThe aim of this study is to investigate sertralin in point of pharmacogenetics and determine the drug and its major metabolite in plasma in depressed patients. For this reason, polymorphisms in genes affecting pharmacokinetic and pharmacodinamic pathways of sertraline metabolism have been investigated in 64 patients who are treated with sertraline. The polymorphisms in those genes were determined by using polymerase chain reaction and restriction fragment length polymorphism. Plasma concentrations of sertraline and its major metabolite desmethylsertraline were measured by using high performance liquid chromatography.en_US
