dc.description.abstractm ÖZET Bu çalışmada önemli bir karsinojen olan DEN verilen ratlarda, lipid peroksidasyonunun son ürünü olan MDA ile SOD ve E vitamini düzeyleri üzerine GSH nın koruyucu etkileri araştırıldı. Kontrol (n=8), DEN verilen grup (n=8), DEN+GSH verilen grup (n=8) olmak üzere 3 grup oluşturuldu. Ratlar 1. 24. 48. 72. 96. saatlerde kanları alındıktan sonra öldürülerek karaciğer ve böbrek dokuları alındı. Plazma MDA düzeylerinde 24. ve 96. saatlerde DEN verilen grupta önemli düzeyde artış gözlendi (p<0.05, p<0.01). Eritrosit SOD aktivİtesinde kontrol grubu ile kıyaslandığında 24. saatte DEN verilen grupta anlamlı düşme (p<0.05), 12. saatte DEN+GSH verilen grupta önemli derecede artış saptandı (p<0.01). E vitamini düzeylerinde DEN verilen grupta 24. ve 72. saatlerde anlamlı azalma vardı (p<0.05). 96. saatte kontrol grubuna göre diğer gruplarda artma vardı (p<0.01). Rat karaciğer dokusunda MDA düzeyleri kontrol grubuna göre diğer gruplarda artmıştı (p<0.05). SOD, DEN verilen grupta azalmıştı (p<0.001). E vitamini DEN+GSH verilen grupta düşmüştü (p<0.05). Böbrek dokusunda kontrol grubuna göre MDA düzeylerinde fark gözlenmezken, SOD aktivitesi DEN verilen grupta azalmış (p<0.01), DEN+GSH verilen grupta artmıştı (p<0.01). Vi+E değerleri ise DEN grubunda artmıştı (p<0.01). DEN'in lipid peroksidasyonu ile SOD ve Vit E üzerine önemli etkisi olduğu, GSH nın bu peroksidasyonda yeterince koruyucu görevi yapmadığı kanısına varıldı. dc.description.abstractIV SUMMARY In this study examined effects of DEN on lipid peroxidation and antioxidants, and also the protective effects of GSH on this mechanism. For this aim, we detarmined the liver and kidney tissue and also erytrocyte SOD activity, vitamin E and MDA levels. The groups were designed as follows: Control (n=8), DEN infused group (n=8), DEN+GSH infused group (n=8). The blood samples were taken at 1, 24, 48, 72, 96th hours after administration and the tissues were remoted immediately. In the DEN group, plasma MDA levels were increased significantly at 24th and 96th hours (p<0.05), p<0.01). The erythrocyte SOD activity was significantly decreased as compared with control at 24th hour in DEN grouh (p<0.05). This enzyme activity was also decreased in DEN + GSH group at 72th hour (p<0.01). Vitamin E levels were decreased in DEN group at 24th and 72th hour (p<0.05). However, vitamin E levels were increased at 96th hour in both two group as compared with controls (p<0.01). Rat liver tissue MDA levels were increased in both two group as compared with controls (p<0.05). The SOD activity was decreased in DEN+GSH group (p<0.05). There was no difference in kidney tissue MDA levels. The SOD activity was decreased in DEN group (p<0.01). However, this enzyme activity was increased in DEN+GSH group (p<0.01). The vitamin E levels was increased in DEN group (p<0.01). In conclusion, DEN has an important effects on lipid peroxidation, SOD activity and vitamin E levels.
