dc.description.abstractÖZETResveratrol (3,4',5 trihydroxystilbene); stilben fitoaleksindir. Resveratrol Polygonum cuspidatum (çoban değneği) bitkisinin kökü, kırmızı şarap ve yer fıstığı yanında, dut, çilek, böğürtlen gibi küçük yumuşak meyvelerde tesbit edilmiştir. Resveratrolün lipid metabolizmasını düzenleme, eikosanoid sentezini inhibe etme ve trombosit çökelmesini baskılama gibi özellikleri yanında antioksidan, antikanserojen, antiinflamatuar, antimutajen, antiproliferatif, antiviral, antibakteriyal, östrojenik ve damar gevşetici özellikler gibi biyolojik aktiviteleri de bulunmaktadır.Taninler moleküler ağırlıkları 500-3000 Da. olan polifenolik bileşiklerdir. Taninler hidrolize ve kondanse taninler olmak üzere iki sınıfa ayrılırlar. Yaygın olarak tannik asit olarak isimlendirilen hidrolize taninler, ya gallotanin ya da ellagiktanin içerirler. Tanin içeren polifenoller anti tümör, antiviral, antiHIV, lipid peroksidasyon inhibisyonu ve plazmin aktivitesi gibi biyolojik aktivitelere sahiptirler.Bu çalışmada, MCF-7 hücre dizisinden oluşturulan gruplarda, resveratrol ve tannik asidin, apoptotik indekse, Bak proteinine ve telomeraz enzim aktivitesine olan etkileri incelenerek bu parametreler üzerine hangi maddenin daha etkili olduğu araştırıldı.Bu çalışmada kullanılan MCF-7 hücre dizisiden, 24., 48. ve 72. saatlerde incelenmek üzere, madde verilmeyen, resveratrol ve tannik asidin 25, 50, 100 µM dozlarının verildiği gruplar oluşturuldu. Bu gruplarda, apoptoz Apop Taq peroxidase kiti (Chemicon) ile Bak proteini [Bak monoklonal antikor (Neomarkers)] immunohistokimyasal boyama yöntemi ile belirlendi. Telomeraz enzim aktivitesi ölçümü ise TeloTAGGG Telomerase PCR ELISA PLUS (Roche) kiti ile belirlendi.Çalışmamızın sonucunda, apoptotik indeks ve % Bak oranını artırmada ve telomeraz enzim aktivitesini düşürmede tannik asit resveratrole göre daha etkili bulundu.Anahtar Sözcükler : Apoptoz, Bak, MCF-7, Resveratrol, Tannik Asit, Telomeraz
dc.description.abstractSUMMARYResveratrol (3,4',5 trihydroxystilbene); stilben is a member of the phytoalexin family. Resveratrol the root of Polygonum cuspidatum was determined in red wine and peanut, as well as in such soft small fruit as mulberry, strawberry and blackberry. Resveratrol is known to regulate metabolism of the lipid, inhibit eicosanoid synthesis and suppress platelet aggregation, apart from functioning as an antioxidant, anti-carcinogenic, anti-inflammatory, anti-mutagenic, anti-proliferative, antiviral, antibacterial, estrogenic and vasodilator agent.Tannins are polyphenolic compounds with a molecular weight of 500-3000 Da. They split into two categories: hydrolysed and condense tannins. Generally known as tannic acid, hydrolysed tannins contain either gallotanin or ellagictanin. Polyphenols containing tannin can function as anti-tumour agents, apart from functioning as antiviral, anti-HIV and inhibiting agent for lipid peroxidation.The present study aimed to investigate effects of resveratrol and tannic acid upon groups made up of MCF-7 cell line, apart from determining their effects upon apoptotic index, Bak protein and telomerase enzyme activities. The present study, therefore, aimed to determine whether resveratrol or tannic acid had more effects upon these parameters.Using substance-free resveratrol and tannic acid with varying doses of 25, 50, 100 µM, groups were established out of MCF-7 cell line to be examined at 24th, 48th and 72nd hours. For these groups, apoptosis was determined via Apop Taq peroxidase kit (Chemicon), while Bak protein [Bak monoclonal antibody (Neomarkers)] was determined via immunohistochemical stain method. Determination of Telomerase enzyme activity was achieved by means of the Telomerase ELISA measurement kit TeloTAGGG Telomerase PCR ELISA PLUS (Roche).The present study concluded that tannic acid was more effective in increasing percentage of Bak protein and decreasing telomerase activity than was Resveratrol.Key words: Apoptosis, Bak, MCF-7, Resveratrol, Tannic Acid, Telomeraseen_US
