dc.description.abstractMiyoglobinürik akut böbrek hasarı, iskelet kas hasarı sonucu gelişen üremik bir sendromdur. Bu çalışmamızda, iskelet kası tarafından üretilen irisinin deneysel miyoglobinürik akut böbrek hasarı modelinin fizyopatolojindeki rolünü araştırmayı amaçladık. Bu çalışmada, 42 sıçan (170-200 gr, erkek, Sprague Dawley) kullanıldı.Kontrol grupları 6 MABH grupları 8 sıçan içeren 3 alt gruba ayrıldı.Tüm sıçanlar 24 saat susuz bırakıldı.kontrol gruplarına saline ve MABH gruplarına %50 gliserol solusyonu intramusküler injekte edildi (8 ml/kg).Her gruptaki sıçanların idrarları toplandı sonra eksanguinasyon yöntemi ile ötenazi yapıldı. Çalışmamızda, K-48 ve MABH-48 ile MABH-24 ve MABH-48 arasında plazma irisin seviyesinde anlamlı artış gösterildi.K-24 ve MABH-24 ile K-48 ve MABH-48 arasında idrar irisin düzeyinde anlamlı azalma bulundu. K-6 ve MABH-6 arasında böbrek doku irisin ekspresyonu arasında anlamlı artış yokken, K-24 ve MABH-24 ile K-48 ve MABH-48 arasında anlamlı bir azalma bulundu. K-6 ve MABH-6, K-24 ve MABH-24, K-48 ve MABH-48, MABH-6 ve MABH-48 ile MABH-24 ve MABH-48 arasında böbrek hasarı skorunda anlamlı bir artış gösterildi. Kontrol ve MABH grupları arasında serum üre, kreatinin, potasyum düzeyleri, ALT, AST, LDH ve CK aktiviteleri karşılaştırıldığında anlamlı bir artış bulundu. Böbrek fonksiyonunda zamana bağlı bozulma ve histopatolojik hasarda artış gösterildi. İdrar irisin seviyesi ve böbrek dokusu irisin ekspresiyonunda azalma olurken, serum irisin seviyesi zamana bağlı olarak artığı gösterildi.Bu bulgular irisinin MABH'nin patogenezinde biyobelirteç olarak kullanılabileceğini desteklemektedir.
dc.description.abstractMyoglobinuric acute kidney injury (MAKI) is an uremic syndrome that develops after a damage in the skeletal muscle cells. In this study, we aimed to investigate role of irisin which produced by the skeletal muscle, on physiopathologyof experimental MAKI. In the study, 42 rats (170-200 g, male, Sprague Dawley) were used. The control group and MAKI group were divided into 3 subgroups. All rats were deprived of water for 24 hours. Control groups were given saline and MAKI groups were injected intramuscular 50% glycerol solution (8ml/kg). The urine samples of the rats in each groups were collected than euthanasia was performed by the exsanguination method. There was a significant increase in plasma irisin levels between C-48 and MAKI-48 groups and between MAKI-24 and MAKI-48 groups (p<0,05). There was a significant decrease in the urine irisin levels between C-24 and MAKI-24 groups and between C-48 and MAKI-48 groups. There was no significant increase in the kidney tissue irisin expression immunohistochemically between C-6 and MAKI-6 groups. On the other hand, there was a significant between decrease C-24 and MAKI-24 groups between C-48 and MAKI-48 groups. For the renal injury score, there was a significant between increase C-6 and MAKI-6 groups, between C-24 and MAKI-24 groups between C-48 and MAKI-48 groups between MAKI-6 and MAKI-24 groups between MAKI-6 and MAKI-48 groups and between MAKI-24 and MAKI-48 groups. There was a significant increase in serum urea, creatinine, potassium levels, ALT, AST, LDH, CK activities between control and MAKI groups (p<0.05).We observed a time-dependent decrease in the renal function and increase in the histopathological damage. While there was a decrease in the urinary irisin levels andkidney tissue irisin expression, serum irisin levels were increased depending on the time basis. These findings support that irisin might be used as a diagnostic marker in the pathogenesis of MAKI.en_US
