dc.description.abstractBu çalışmada sıçanlarda deneysel stres modeli ile indüklenen gastrik lezyonlara karşı probiyotik Lactobacillus rhamnosus GG (ATCC 53103)' nin koruyucu etkileri araştırılmıştır. Bu çalışmada sağlıklı, 8-9 aylık ve ortalama 420 gr ağırlığında Wistar albino erkek sıçanlar arasından rastgele seçimle her birinde n=7'şer sıçan olmak üzere toplam 6 grupta toplam 42 adet sıçan kullanılmıştır. Mide dokusunda ülserleşme oranı stres grubunda yüksek oranda (83.5±18.42 mm2) bulunurken M5 grubunda (5.03±4.62 mm2) kontrol grubuna yakın değerde bulundu. Stres grubundaki asidik pH değerini (3.26±0.71) M5 grubunda Lactobacillus rhamnosus GG bakterisinin azalttığı (4.75±1.60) görülmüştür. Serumda glukokortikoid (GK) seviyesi stres grubunda (45.76±14.50 ng/L) pantoprazol grubuna göre (26.47±4.84 ng/L) önemli düzeyde yüksek oranda görülmüştür. Mide dokusunda katalaz (KAT), süperoksit dismutaz (SOD) ve malondialdehit (MDA) seviyelerinde kontrol grubuna göre önemli bir değişiklik gözlenmemiştir. Glutatyon peroksidaz (GSH-Px) seviyesinin pantoprazol grubuna (50.04±9.21 U/ml) göre stres (76.52±10.62 U/ml) ve M5 grubunda (76.54±20.37 U/ml) önemli düzeyde yüksek oranda olduğu görülmüştür. Nitrik oksit (NO) seviyesinin M5 grubunda (43.01±8.76 µmol/L) pantoprazol grubuna (24.08±1.21 µmol/L) göre önemli düzeyde yüksek oranda olduğu görülmüştür. Midenin epitel dokusunda yoğun hücresel hasar stres grubunda bulunurken kontrol grubuna kıyasla midenin epitel dokusunda en az hücresel hasar M5 grubunda görülmüştür. Kontrol grubu ve M5 grubuna ait mide preparatlarında benzer oranda mast hücreleri izlenirken, stres grubunda, kontrol grubu ve M5 grubuna oranla daha fazla sayıda mast hücresi izlenmiştir.Anahtar kelimeler:Stres, ülser, probiyotik, laktobasillus, oksidatif stres, antioksidan
dc.description.abstractIn this study, protective effects of probiotic Lactobacillus rhamnosus GG (ATCC 53103) were investigated against gastric lesions by induced experimental stress model in rats. In this study, Wistar albino male rats were used and they were 8-9 mounths old and had 420 gr weight. Each group had 7 rats and totaly 42 rats used in 6 groups. In the stomach tissue, ulceration rate was found in high level in stress group (83.5±18.42 mm2) and was found similar to control group in M5 group (5.03±4.62 mm2). Lactobacillus rhamnosus GG decreased the acidic pH level of the stress group (3.26±0.71) in the M5 group (4.75±1.60). In the serum, glucocorticoid level was found at a significant level in the serum group (45.76±14.50 ng/L) when compared to the pantoprazole group. In the stomach tissue in levels of catalase (CAT), superoxide dismutase (SOD) and malondialdehyde (MDA) was not observed a significant change when compared to the control group. Glutathione peroxidase (GSH-Px) level was found at a significant level in the serum group (76.52±10.62 U/ml) and M5 group (76.54±20.37 U/ml) when compared to the pantoprazole group (50.04±9.21 U/ml). Nitric oxide (NO) level was found at a significant level in the M5 group (43.01±8.76 µmol/L) when compared to the pantoprazole group (24.08±1.21 µmol/L). The most intensive cellular damage in gastric epithelial tissue was found in the stress group and minimum cellular damage in gastric epithelial tissue was observed in the M5 group. In control and M5 groups, mast cells were observed at the similar rate. When compared to the control group and the M5 group, more much mast cells were observed in the stress group, pantoprazole group, M1, and especially M10 group.Key words:Stress, ulcer, probiotic, lactobacillus, oxidative stress, antioxidanten_US
