dc.description.abstractÇalışmamızda asetik asit ile deneysel kolit oluşturulan sıçanlarda, sirolimusun inflamatuvar sürece etkisini, makroskopik, ışık mikroskopik, biyokimyasal ve immünohistokimyasal olarak araştırdık. Otuz adet dişi Spraque?Dawley sıçan 10?arlı 3 gruba ayrıldı: 1. grup kontrol grubu, 2. grup kolit grubu, 3. grup kolit oluşumuyla eş zamanlı sirolimus tedavisi verilen grup.Makroskopik ve mikroskopik olarak; kolit oluşturulan gruplar arasında istatistiksel yönden anlamlı bir fark saptanmadı (p>0,05). Kolit grubunda interlökin-1 ve interlökin-10 düzeylerinin belirgin olarak arttığı, kolit oluşturulup sirolimus verilen grupta ise kolit grubuna göre interlökin-1 ve interlökin-10 düzeylerinin istatistiksel açıdan anlamlı düşük olduğu tespit edildi (p<0,05). Kolit grubunda cluster of differentiation 3+, cluster of differentiation 5+ ve cluster of differentiation 45+ T lenfositlerinin dağılım ortalamasının belirgin olarak arttığı görüldü. Kolit oluşturulup sirolimus verilen grupta prolifere hücre nükleer antijeni proliferasyon indeksi ortalamasının istatistiksel açıdan kolit grubuna göre yüksek olduğu saptandı (p<0.05).Sonuç olarak; inflamatuvar barsak hastalığının tedavisinde sirolimusun etkinliğini araştıran daha geniş kapsamlı çalışmalara ihtiyaç olduğunu düşünüyoruz. dc.description.abstractIn our study, we investigated the effect of sirolimus on inflammatory processes by macroscopic, light microscopic, biochemical and immunohistochemical evaluation in a experimental colitis rat model with acetic acid. Thirty female Sprague-Dawley rats were divided into 3 groups, each group consisting 10 animals. First group was the control group, second group was colitis group and the third group was colitis group which was treated with sirolimus.There was no statistically significant difference found between the colitis groups in macroscopic and microscopic evaluation (p> 0,05).Interleukin-1 and interleukin-10 levels were significantly increased in colitis group. However there was a statistically significant decrease in interleukin-1 and interleukin-10 levels in colitis group which was treated with sirolimus(p <0,05). Cluster of differentiation 3+, cluster of differentiation 5+ and cluster of differentiation 45+ T lymphocytes average range was were significantly increased in colitis group. Proliferating cell nuclear antigen proliferation index in colitis group which was treated with sirolimus was statistically higher than the colitis group (p <0.05).As a result, more extensive studies should be performed about the effects of sirolimus in the treatment of inflammatory bowel disease.Key words: Ulcerative colitis, inflammatory bowel disease, rapamycinen_US
