dc.description.abstractÖZET Bu çalışmada, Wistar Albino sıçanlara gentamisin kg başına günde 5,40 ve 80 mg olmak üzere oniki gün süre ile uygulandı. Uygulanan doza ve zaman sürecine bağlı olarak gentamisin'in nefrotoksik etkisi biyokimyasal yönden değerlendirildi. Serum örneklerinde; üre azotu, kreatinin, ifosfat, ürik asit, LDH ve GST deney gruplarında değişen düzeylerde yüksek, kreatinin klirens düşük bulunurken (P<0.05, P<0.01, P<0.001), T.protein düzeyleri değişmedi (P>0.05). Kalsiyum, albumin, ALP ve GGT, G-5 grubunda farksız bulundu (P>0.05); G-40 G-80 gruplannda ise kalsiyum ve albumin düzeylerinin düştüğü (P<0.05), ALP ve GGT aktivitelerinin yükseldiği gözlendi (P<0.01, P<0.001). GGT, G-40 ve G-80 gruplannda (P<0.05, P<0.001), ALP, LDH ve GST deney gruplarının her birinde düşük (P<0.05, P<0.01, P<0.001) bulunmuş, ilacm doku harabiyeti oluşturduğu kanısına vanlmıştır. G-5 grubu idrar örneklerinde; 4. günde sadece GST salınımı saptanırken (P<0.05), üre azotu, kreatinin, ürik asit düzeylerinde düşme ve protein, GGT, LDH atılımında artış 8. günde başladı (P<0.05), 12. günde dansite yükselirken, Î.fosfat düzeyinde düşme gözlendi (P<0.05). Deney sonunda, hacim, pH, kalsiyum ve ALP farksız bulundu (P>0.05). G-40 grubu idrar örneklerinde; 4. günde dansite, üre azotu, kreatinin, î.fosfat ve ürik asit düzeylerinde düşme, ALP, GGT, LDH ve GST atılımında artış hızla başladı (P<0.01). Protein ve pH 8. günde artış gösterdi (P<0.01). 12 günde pH normale dönerken (P>0.05), hacimde azalma gözlendi (P<0.01). G-80 grubu idrar örneklerinde; hacim 8. günden, diğer parametreler 4* günden, itibaren ileri düzeyde farklı bulundu (P<0.001). İncelemelerimize dayanarak, idrar enzim aktivitelerinin tubuler harabiyeti gösterilebileceği, özellikle GST'nin erken dönemde tanıya yardımcı olabileceği sonucuna varıldı.
dc.description.abstractIV SUMMARY In this study gen tamirine was applied to Wistar Albino rats 5,40 and 80 mg/kg per day for 12 days. The effect of nephrotoxicity of gentamicine was evaluated biochemically depending on the dose applied and the duration of time. While, in sera samples, urea nitrogen, creatinine, I.phosphate, uric acid, LDH and GST were found high in varying levels (P<0.05, P<0.01, P<0.001), and creatinine clearance was found low (P<0.05, P<0,01, P<0.001), T.protein levels did not change (P>0.05). Calcium, albumin, ALP and GGT, were found without any difference in G-5 group (P>0.05), and it was observed that calcium and albumin levels decreased in G-40 and G-80 groups (P<0.05), ALP and GGT activities increased (P<0.01, P<0.001). In tissue samples, GGT, G-40 and G-80 (P<0.05, P<0.01) groups, ALP, LDH and GST in each of experimental groups were found low (p<0.05, P<0.01, P<0.001), thus, the medicine was throught to cause tissue damage. While GST extraction were determined alone during the 4th day in the urine samples of G-5 group (P<0.05), a decrease started in the levels of urea nitrogen, creatinine and uric acid; an in crease started in the extraction of protein, GGT, and LDH during the 8th day (P<0.05). We observed an increase in density during the 12th day, and a decrease in Lphosphate level (P<0.05). At the end of the experiment, the volume, pH, calcium and ALP were found without any difference (P>0.05). In G-40 group urine samples during the 4th day, a decrease started in density, urea nitrogen, creatinine, I.phosphate and uric acid and a fast increase started in the extraction of ALP, GGT, LDH and GST (P<0.01). Protein and pH tended to increase during the 8th day. While pH normalized during the 12* day (P>0.05, a decrease was observed in volume (P<0.01). In the urine samples of G-80 group volume was found very much different after the 8th day, and the other parameters were found very much different after the 4th day (P<0.001). As a result of our studies it has been concluded that enzyme activities can indicate tubular damage, and especially GTS can be helpful in diagnosing at an early stage.en_US
