dc.description.abstractBu çalismada siçan beyin mikrodiyalizatlarinda bazi nörokimyasal maddelerintayini için elektrosprey iyonizasyonlu sivi kromatografi kütle spektrometri (ESILC-MS) yöntemi tanitilmaktadir. Iyon tuzagi kütle analizörü ile sekiz adetnörokimyasal (epinefrin, norepinefrin, dopamin, 3,4-dihidroksifenilasetik asit(DOPAC), histamin, g-aminobutirik asit, L-glutamat, asetilkolin) için kütlespektrumlari ve kütle parçalanmalari belirlendikten sonra sivi kromatografikayirma kosullari incelenmistir. C18 kolon (2.1x100 mm, 3.5 ?m) ve % 0.1 asetikasit içeren su-asetonitril gradient hareketli faz sisteminde türevlendirici ajan olan4-kloro-7-nitro-2,1,3-benzoksadiazol (NBD-Cl) ile türevlendirilerek ayrilan g-aminobutirik asit ve histamin için yöntemin validasyonu gösterilmistir. GABANBDiçin 267à249 m/z kütlesinden olusan parçalanma kütlesi, histamin içinsadece 275 m/z ana kütlesi kullanilarak olusturulan kromatogramda miktar tayiniyapilmistir. Dogrusal aralik g-aminobutirik asit için 10-500 ng.mL-1, histamin için50-1000 ng.mL-1 bulunmustur. Yöntemin tayin sinirlari g-aminobutirik asit için 10ng.mL-1, histamin için 50 ng.mL-1 olarak hesaplanmistir. Yöntemin GABA vehistamin için elde edilen dogrulugu % 98 dolayinda saglanan geri kazanim ile,tekrar edilebilirligi ise gün içi % 1.65-2.62, günler arasi % 1.23-4.93 bagilstandart sapma degerleri ile gösterilmistir. Siçan beyninin ventral anteriornucleus, lateral hypothalamus ve corpus striatum bölgelerinden toplananmikrodiyalizat örneklerinde g-aminobutirik asitin bazal düzeyleri ölçülereksirasiyla 123.25±0.67, 151.41±18.53, 94.96±0.53 ng.mL-1 olarak bulunmustur.Anahtar Kelimeler: Nörokimyasallar, Histamin, g-Aminobutirik Asit,Türevlendirme, Mikrodiyaliz, LC-MS
dc.description.abstractAn electrospray ionization-liquid chromatography-mass spectrometry (ESI-LCMS)method is applied to the simultaneous determination of certainneurochemicals in rat brain microdialysate. The separation condition wasexamined for liquid chromatography after evaluation of mass spectra and massfragments for eight neurochemicals (epinephrine, norepinephrine, dopamine,DOPAC, histamine, g-aminobutiric acid, L-glutamate, acetylcholine) by ion trapmass analyser. The method has been validated using C18 column (2.1x100 mm,3.5 ?m) and gradient mobile phase containing 0.1 % glacial acetic acid in wateracetonitrileafter derivatization of his tamine and g-aminobutiric acid with afluorescent derivatization reagent of 4-chloro-7-nitro-2,1,3-benzoxadiazole (NBDCl).Characteristic precursor-to-product ion transitions, m/z 267à249 for GABANBDand only m/z 275 for histamin-NBD were monitored for the quantification.Linearity range was 10-500 ng.mL-1 for g-aminobutiric acid and 50-1000 ng.mL-1for histamine. The limit of quantification was calculated as 10 ng.mL-1 and 50ng.mL-1 for g-aminobutiric acid and histamine, respectively. Accuracy was shown98 % method recovery for histamin and GABA. The method precision was shownby day to day repeatability and intermediate precision found as 1.65-2.62 % RSDand % 1.23-4.93 % RSD, respectively. Microdialysate sampling for g-aminobutiric acid and histamine was performed in ventral anterior nucleus,lateral hypothalamus and corpus striatum of rat brain and basal levels of g-aminobutiric acid were determined as 123.25±0.67, 151.41±18.53, 94.96±0.53ng.mL-1 respectively.Key Words: Neurochemicals, Histamine, g-Aminobutiric Acid, Derivatization,Microdialysis, LC-MSen_US
