dc.description.abstract- 48 - VL ÖZET Bu çalışmada kolon kanserli hastaların tümörlü ve nor mal kolon dokusundaki protein dağılımları incelendi. 1-8 nolu olguların kolon materyalinin normal ve tümörlü kısımlar ayrılarak perklorik asit presipitasyonu ile 9-10 nolu olgular ise amonyum sülfat presipitasyonu ile protein fraksiyonları elde edildi. 6 olguda (1-3-5-8-9-10) kolon dokusunun normal kısmında total protein miktarı kanserli dokuya göre daha fazla bulundu. 3 olguda (4-6-7) ise kanserli kısımda total protein miktarında artış gözlendi. 1-8 olguların protein de ğerleri istatistiksel olarak değerlendirildiğinde normal kı sımda 0.085±0.034 ug/g doku, tümörlü kısımda 0.087 ± 0.033 ug/g doku olarak bulundu. Çöktürülen protein fraksiyonlarında PAGE sonuçlarında tümörlü kolon materyalinde 26.000, 54.000, 55.000, 56.000, 64.000, 66.000 ve 96.000 molekül ağırlıklı proteinler görülürken normal kolon materyalinde 24.000, 51.000, 68.000, 72.000 molekül ağırlıklı proteinlerin varlı ğı görüldü. Bir olgu haricinde bütün örneklerde tek protein bandı görülürken genelde normal kolon veya tümörlü kolon ma teryalinde ortak bir bant elde edilemedi. Fakat her bir olgu kendi içinde tümörlü ve normal kolon dokusu olarak mütalaa edildiğinde farklılıklar gözlendi. Bu bulgular her kolon kan seri dokusuna spesifik bir proteinin olabileceğini düşündür mektedir. 9-10 nolu olgularda amonyum sülfat presipitasyonu ile elde edilen, normal ve tümörlü kolonun protein fraksiyon larına ve 9. ve 10. olgularda CEA düzeylerine bakıldı.49 - The colon protein distribution in colon cancer patients. VIL SUMMARY In this work the protein distribution was examined in the tumoral and normal parts of the colon tissue of colon cancer patients. The protein fractions were isolated either by perchloric acid precipitation (Cases:l-8) or by ammonium sulphate precipitation (Cases:9,10) of the tumoral and normal parts of the colon materials. In six of the cases (Case No:l,.3,5,8,9,10) the protein concentrations were higher in the normal parts compared to tumoral parts whereas in the three cases (Case No:4,6,7) the protein concentrations were higher in the tumoral parts. But when the protein concentrations were analyzed statistically in the 8 cases (Case No:l-8) together no difference was observed where the protein concentration values were 0.085±0.034 yg/g and 0.087±0.033 ug/g in normal parts and tumoral parts, respectively. These protein fractions were analyzed by SDS-PAGE. In the tumoral colon materials the proteins with molecular weights of 26.000, 54.000, 55.000, 56.000, 64.000, 66.000 and 96.000 and in the normal parts 24.000, 51.000, 68.000, 72.000 Da proteins were detected. Except in one case in all of the samples only one protein band was observed where no common band between the cases or between the tumoral and normal was observed. Also CEA levels were determined and CEA was detected both in the normal and cancerous parts. The protein fractions of the cases 9 and 10 were also analyzed with isocratic HPLC. Protein peaks with Rts 11:50 and 12:00 were observed in the tumoral and normal samples, r ecpectively.
