dc.description.abstractÖZET : Vücut sıvılarında poliamin düzeylerinin artmasının hücre proliferasyonunun artması ve dokuların hızla çoğalması olgularıyla bağlantılı olduğu bilinmektedir. Poliarninler kanser vakalarının tedavüerinin düzenlenmesinde yararlanılan duyarlı markerler olarak değerlendirilebilir. Akciğer,over ve meme kanserli vakalarda serum poliamin (putresine- put, kadaverine-cad, spermidine-spd, spermine-spn) ve diğer tümör marker ( CEA, CA125, CA19-9, CA 15-3, Sialik Asit) düzeyleri bu çalışmada incelenmiştir. Bu çalışmada gradiyent HPLC ile yapılan pohamin analiz yöntemi geliştirilmiştir. Diğer tümör markerlerinin analizi de immünolojik ve kolorimetrik yöntemlerle yapılmıştır.. Kontrol grubu. (n= 18, kanser olmayan kişiler) serum poliamin ve tümör markerlerin ortalama değerleri aşağıdaki gibidir: put : 0.73±0.56 nmol/ml, cad : 0.19±0.24 nmol/ml, spd : 0. 3 9±0. 41 nmol/ml, spn : 0.17±0.17 nmol/ml, CA19-9 : 16.92±6.93 U/ml, CA125 : 13.08±6.43 U/ml, CA15-3 : 18.67±5.82 u/ml CEA : 2.73±1.66 ng/ml, Sialik Asit : 1.81±0.26 nmol/ml Klinik olarak progresyondaki akciğer kanserli vaka grubu(n=21) ortalama değerleri : put : 14.83±10.60 nmol/ml, cad: 4.25±6.20 nmol/ml spd : 2.79±4.1 1 nmol/ml, spn : 0.22±0.19 nmol/ml,CA 19-9 : 157.81±129.25 u/ml, CEA : 42.50±63.27 ng/ml, Sialik Asit : 3.58±0.87 umol/ml. 85Remisyondaki akciğer kanserli vakaların (n=20)ortalamala değerleri: put : 0.89±0.57 nmol/ml,cad: 0.58±0.52 nmol/ml,spd: 0.28±0.34nmol/ml, spn : 0.14 ±0.14nmol/ml, CA19-9 : 18.69±9.12 U/ml CEA : 4.16±2.34 ng/ml, Sialik Asit : 2.37±0.51 umol/ml Progresy ondaki over kanserli vakaların (n=16) ortalama değerleri: put : 9.92±11.76 nmol/ml, cad : 9.12±8.64 nmol/ml, spd : 3.15±5.76nmol/ml, spn : 0.1 6±0. 14 nmol/ml, - CA125 : 158.63±145.58 U/ml,Sialik Asit : 3.33±0.89 umol/ml Remisyondaki over kanserli vakaların (n=18)ortalama değerleri: put : 0.61±0.42 nmol/ml,cad : 0.39 ±0.40nmol/ml,spd : 0.36±0.29 nmol/ml spn : 0.18±0.80 nmol/ml, CA125 :12.83 ±10.80 U/ml Sialik Asit :2.68±0.68 umol/ml Akciğer ve over kanserli vakalarda putresin ve kadaverin düzeyleri kontrol grubuna kıyasla anlamlı artışlar göstermiştir.Her iki poliamin düzeyleri progresy ondaki vakalarda artmakta ve remisyona giren vakalarda normal düzeylere dönmektedir. Spermidin düzeylerinde de paralel bir-artış görülmesine rağmen sekonder bir önem taşımaktadır. Meme kanserli vakalarda da serum poliamin düzeylerinin anlamlı artış göstermediği saptanmıştır...Bu çalışmada elde edilen bulgular putresin ve kadaverinin akciğer ve over kanserlerinde spesifik prognostik tümör marker olarak değerlendirilebileceklerini göstermektedir. 86
dc.description.abstractSUMMARY: Elevation of polyamines in body fluids has been reported to be associated with increased cell proliferation and correlated with rapidly growing tissue. Polyamines may be sensitive markers for monitoring therapy of cancer patients. The level of polyamines (putrescine-put, cadaverine-cad, spermidine- spd, spermine-spn) and other tumor markers ( CEA, CA125, CA19-9,CA 15-3, Sialic Acid) were evaluated in the serum of lung, ovary, and breast cancer patients at the present study Progression and remission groups were compared to normal group. Gradient HPLC analysis were performed for polyamine determinations. Other tumor markers was determined by immunological and colorimetric methods. In the control group(n=18, healthy subjects) serum polyamine and tumor marker mean values were:put : 0.73±0.56 nmol/ml, cad: 0.19±0.24 nmol/ml, spd : 0.3940.41nmol/ml,spn : 0.17±0.17 nmol/ml, CA19-9 : 16.92±6.93U/ml, CA125 : 13.08±6.43 U/ml, CA15-3 : 18.67±5.82 U/ml CEA:2.73±1.66ng/ml, Sialic Acid : 1.81±0.26 umol/ml 87Clinically progressing lung cancer patients (n=21) had serum polyamine mean values ± SD as follows: put : 14.83Ü0.60 nmol/ml, cad : 4.25±6.20 nmol/ml spd : 2.79±4.11nmol/ml, spn : 0.22±0.19 nmol/ml,CA 19-9 : 157.81±129.25 U/ml, CEA : 42.50±63.27 ng/ml, Sialic Acid : 3.58±0.87 umol/ml. Lung cancer patients in remission(n=20) had mean values ±SD as follows: put : 0.89±0.57 nmol/ml,cad : 0.58±0.52 nmol/ml,spd : 0.28=t0.34nmol/ml, spn: 0.14 ±0.14nmol/ml, CA19-9 : 18.69±9.12 U/ml CEA : 4.16±2.34 ng/ml, Sialic Acid : 2.37±0.51 umol/ml Ovary cancer patients with progressing disease(n=16)had mean values ±SD as follows: put : 9.92Ü1.76 nmol/ml, cad : 9.12±8.64 nmol/ml, spd : 3.15±5.76nmol/ml, spn : 0.16±0.14 nmol/ml, CA125 : 158.63±145.58 U/ml,Sialic Acid :3.33±0.89 umol/ml Ovary cancer patient in remission(n= 18) had mean values ±SD as follows: put : 0.61±0.42 nmol/ml,cad : 0.39 ±0.40nmol/ml,spd : 0.36±0.29 nmol/ml spn : 0.18±0.80 nmol/ml, CA125: 12.83 ±10.80 U/ml Sialic Acid: 2.68±0.68 umol/ml Putrescine and cadaverine serum levels in lung and ovary cancer patients are significantly elevated compare to the control group. Both parameters show elevation in progressing cases and in remission the values were found as in normal group. Spermidine levels show parallelism but have secondary importance. Increase in serum polyamine levels in mammary cancer patients were insignificant. Present data indicates that evaluation of serum putrescine and cadaverine levels could be specific prognostic tumor markers for lung and ovary cancer patients. 88en_US
