dc.description.abstractIn studies of hybridoma cell and vaccine preparation, in order to obtain a specificantibody, the selection of adjuvants, especially those which may help to increase theantigenicity of a particular substance is essential. In this perspective ,in the study presentedthe use of polyelectrolyte adjuvants was investigated.For the development of a model system, Bovine serum albumine (BSA) was complexedwith a number of different sytnthetic polymers which posses different physicochemicalproperties. Negatively charged polyacrilic acid (P AA) and BSA molecules were ligated byCu2+ions and the light -scattering method stable complexes and conjugates, which wereobserved soluble or insoluble forms in water and their immunological properties wereanalysed. The results obtained from the analysis of the correlation between thephysicochemical properties of polycomplexes and their immunological activities in vivosystems were as follows. (1) When polyanions were complex with BSA, the immunoresponse was increased by 4 to 5 fold, (2) this increase could only be observed only whenthe three substances, Cu2+, BSA and PAA were all participated in the complexes formed,and (3) it was found that antigens that were complexed with polymers required muchshorter time of immunization than that of conventional immunization procedures.Based on the resuls obtained from the model system described above, the second part ofthe thesis was concerned with the immunological studies on BSA -Progesterone (BSAP)complexes and the complexes they formed with polymer adjuvants. When compared to theresuls of controls used, immuno responses specific to progesterone appeared to beconsiderably higher. A comparative study of immunologic properties of polycomplexesversus BSAP-mcomplete Freund's adjuvant (IF A) mixtures revealed specific differences inthe antibody production. The immunized mice with BSAP+ polycomplexes and BSAP+IF A were used for fusion. 12 monoclonal antibodies of IgG 1 and IgG2a types wereobtained and the affinities of these antibodies were found to be 10.7_10.8 M . There was nocross reaction with the other steroid hormones.In consideration of the whole data obtained, it can be suggested that a helpful method forthe increase of the immunogenicity of antigens with little or no immunogenicity wasdescribed.en_US
