dc.description.abstractiiÖZETBu çalisma, Isparta yöresindeki domates üretim alanlarinda bulunan viralenfeksiyonun belirlenmesi amaciyla 2001-2003 yillari arasinda yürütülmüstür.Üretim alanlarinda yapilan sürveyler sirasinda bitkilerde hiyar mozayik virüsününtipik simptomlarinin gözlenmesi arastirmayi bu dogrultuda yönlendirmistir.Tanilama çalismalarinda mekanik inokulasyon, DAS-ELISA testleri ve dsRNAanalizi yapilmistir.Sürveyler sirasinda toplanan yaprak örneklerinin, hassas test bitkilerine mekanikinokulasyonlari sonucunda hiyar mozayik virüsü için tipik belirtiler gözlenmistir.Viral enfeksiyon belirtisi gösteren bitkilerden alinan toplam 138 örnek DAS-ELISAyöntemine göre test edilmis ve bu örneklerden 56 adedinin CMV ile enfekteli oldugusaptanmistir. Alinan örneklerin % 40.57'sinde CMV enfeksiyonu bulunmustur.CMV'nin dsRNA izolasyonu ve analizi çalismalarinda, çarpici sistemik enfeksiyonsergileyen Nicotiana tabacum ?Xanthii?, Nicotiana tabacum ?Samsun NN? veCapsicum annuum L. bitkilerinin inokulasyondan 12 gün sonra hasat edilen 7'ser grenfekteli yaprak dokulari kullanilmistir. Virüse ait dsRNA'lar %1.2'lik agar jelde100 V.'da 2.5-3 saat süreyle elektroforetik ayrima tabi tutulduklarinda CMV'ye aitüç banta sahip dsRNA profilleri elde edilmistir. Nicotiana tabacum ?Xanthii? veCapsicum annuum L.'den elde edilen profiller Nicotiana tabacum ?Samsun NN?'denelde edilen profilden daha belirgin oldugu görülmüstürAnahtar Kelimeler: Domates, Hiyar mozayik virüsü, ELISA, dsRNA Analizi.
dc.description.abstractiiiABSTRACTThis study was conducted between 2001 and 2003 to determine viral pathogens intomato production areas in Isparta province. Due to high occurrence of cucumbermosaic virus (CMV) symptoms in field surveys the study mainly focused onidentification of this virus in tomato plantations.For this purpose, different identification methods including mechanical inoculationto indicator plants, DAS-ELISA and dsRNA analysis were used in this study. Virus-infected samples collected during surveys were first mechanically inoculated intosensitive indicator plants for symptom development. These plants produced typicalCMV symptoms suggesting that field samples were infected with CMV.Then 138 infected field samples were analyzed by DAS-ELISA. DAS-ELISAshowed that among 138 samples, 56 were infected with CMV indicating that about40.57% of the field samples were infected with CMV.Later, 7 gr leaf samples were collected from systemically CMV infected Nicotianatabacum ?Xanthii?, Nicotiana tabacum ?Samsun NN? and Capsicum annuum L.plants 12 days after inoculation for dsRNA isolation.Finally, isolated dsRNA were analyzed in 1.2% agarose gel by electrophoresis at 100volt for 2.5-3 hr. dsRNA analysis revealed the existence of CMV specific dsRNAprofile in plants confirming the results of previous assays. DsRNA profiles observedin infected Nicotiana tabacum ?Xanthii?and Capsicum annuum L. were moreobvious then that of Nicotiana tabacum ?Samsun NN?.Key Words: Lycopersicum esculentum, cucumber mosaic virus, ELISA, dsRNAAnalysis.en_US
