dc.description.abstractiiÖZETTetrasiklinler veterinerlikte hastaliklarin önlenmesi ve kontrol edilmesi amaciylarutin olarak kullanilmaktadirlar. Genis antibakteriyel spektrumlu olan vebakteriyostatik aktiviteye sahip bu antibiyotikler, Gram-positif and Gram- negatifbakterilerin sebep oldugu hastaliklara karsi etkilidirler. Bununla birlikte, bu ilaçlarinkullanilmasi bulasici hastaliklar gibi ciddi problemlere yol açabilmektedir. Bumaddelerin sütteki kalintilari toksik etki ya da baska alerjik reaksiyonlara sebepolabilmektedir.LC, sütte bulunan tetrasiklinlerin tayini için oldukça yaygin olarak kullanilan biryöntemdir. Bu çalismada, tetrasiklinlerin kromatografik ayirimi, C8 ters- faz kolonda,25mM fosforik asit içeren asetonitril-su mobil fazi kullanilarak gerçeklestirilmistir.Bu çalismada, mobil faz pH'sinin kromatografik ayirmaya etkisi incelenmistir.Bu çalismanin amaci, TC, OTC, MC, DC ve CTC'nin ayirimi için spesifik, hizli vebasit bir yöntem gelistirmektir. Çalismada doksisiklinin kantitatif tayininde, diskalibrasyon grafigi kullanilmistir. dc.description.abstractiiiABSTRACTTetracyclines are used routinely in veterinary medicine for prevention and control ofanimal diseases. They are antibiotics with a broad antibacterial spectrum andbacteriostatic activity and have a good activity against acute disease caused byGram-positive and Gram-negative bacteria. However, the use of these drugs hasbecome a serious problem as regards infectious diseases, as they are substances thatleave residues in milk, which can be directly toxic or else cause allergic reactions.LC is the chromatographic approach most commonly used for the determination oftetracyclines in milk. Chromatographic separation of the tetracyclines was achievedon a C8 reversed-phase column, using a consisting of acetonitrile-water (25:75 v/v)including 25mM phosphoric acid. In this study, the effect of mobile phase pH on thechromatographic separation was investigated.The aim of this work to develop spesific, rapid and simple multiresidue method forseparation of TC, OTC, MC, DC and CTC. In the study quantitative determination ofDC was investigated with external calibration plot. Linear calibration plot wasobtained with correlation coefficient 0,999.Key words : Tetracyclines, LC, dissociation constant, pH optimization in mobilephaseen_US
