dc.description.abstractAlopesi areata (AA) etyolojisi kesin olarak bilinmeyen fakat organ spesifik ototimmün hastalık olarak kabul edilen bir durumdur. Oksidatif stres, oksidan-antioksidan mekanizmalar arasındaki dengesizliktir. Bu çalışmada AA patogenezinde oksidatif stresin rolünü ve hastalık şiddeti, süresi, tekrarı ve paterni ile ilişkisini araştırdık.Çalışmaya 62 hasta ve yaş-cinsiyet uyumlu 62 kontrol alındı. Hastalık şiddeti SALT (Severity of Alopecia Tool) skoru kullanılarak tanımlandı. Hastalar hastalık şiddeti, süresi, tekrarı ve paterni açısından gruplara ayrıldı. Hasta ve kontrol grubunun alınan kan örneklerinde plazma malondialdehid (MDA) seviyesi, eritrosit katalaz (CAT), glutatyon peroksidaz (GSH-Px) ve süperoksid dismutaz (SOD) aktiviteleri ölçüldü.Hastaların 24'ü erkek, 38'i kadın olup, yaş ortalaması 23,7±11 idi. Eritrosit GSH-Px ve SOD aktiviteleri hastalarda kontrol grubuna göre istatistiksel olarak anlamlı oranda düşük (p=0,00 ve p=0,00), plazma MDA seviyesi istatistiksel olarak anlamlı olmamakla birlikte yüksek (p=0,07) ve eritrosit CAT aktivitesi aynı olarak (p=0,20) saptandı. Hastalık şiddeti ile plazma MDA seviyesi (p=0,192), eritrosit CAT (p=0,144), GSH-Px (p=0,183) ve SOD (p=0,395) aktiviteleri arasında ilişki saptanmadı. Hastalık süresi ile plazma MDA seviyesi (p=0,125), eritrosit CAT (p=0,449), GSH-Px (p=0,750) ve SOD (p=0,413) aktiviteleri arasında ilişki saptanmadı. Hastalık tekrar sayısı ile plazma MDA seviyesi (p=0,249), eritrosit CAT (p=0,822), GSH-Px (p=0,561) ve SOD (p=0,447) aktiviteleri arasında ilişki saptanmadı. Hastalık paterni yönünden yama ve yama+ofiyazik paternleri arasında plazma MDA seviyesi (p=0,119), eritrosit CAT (p=0,434), GSH-Px (p=0,375) ve SOD (p=0,146) aktiviteleri açasından istatistiksel olarak anlamlı fark saptanmadı.Sonuç olarak; AA'lı hastaların eritrosit GSH-Px ve SOD aktiviteleri azalmış, plazma MDA seviyesi artmış, eritrosit CAT aktivitesi ise kontrol grubu ile aynı olarak saptandı. Çalışmamızda MDA seviyesindeki artış AA'lı hastalarda oksidatif stresi gösterdi. SOD ve GSH-Px aktivitesindeki azalmanın süperoksid radikallerindeki artış sonucunda olabileceği ileri sürülebilir.
dc.description.abstractThe pathophysiology of alopecia areata (AA) has not been clearly defined. However, it is known as a tissue-restricted autoimmune disease. Oxidative stress arise as a result of disequilibrium between oxidant and antioxidant systems. The aim of this study is to investigate the role of oxidative stress in the etiopathogenesis of AA and evaluation of relationship between oxidative stres and the severity, period, recurrence and pattern of disease.The study included 62 patients with AA. Age and sex matched 62 healthy individuals served as control group. SALT (Severity of Alopecia Tool) score was used for defining severity of disease. Patients were divided into groups according to severity, period, recurrence and pattern of disease. Blood samples were taken from patients and controls. The level of serum malondialdehyde (MDA), activities of erythrocyte catalase (CAT), glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) were measured in this samples.Twenty-four of the patients were male and 38 patients were female, and mean age of all patients was 23,7±11 years. The erythrocyte activity of GSH-Px and SOD in patients with AA were significantly lower than those of controls (p=0,00, p=0,00). The plasma level of MDA in patients with AA were insignificantly higher than controls (p=0,07). No significant difference was observed between the patient and control groups for erythrocyte CAT activity (p=0,20). There was no relationship between severity of disease and plasma level of MDA (p=0,192), erythrocyte activities of CAT (p=0,144), GSH-Px (p=0,183) and SOD (p=0,395). There was no relationship between the period of disease and plasma level of MDA (p=0,125), erythrocyte activities of CAT (p=0,449), GSH-Px (p=0,750) and SOD (p=0,413). There was no relationship between the recurrence of disease and plasma level of MDA (p=0,249), erythrocyte activities of CAT (p=0,822), GSH-Px (p=0,561) and SOD (p=0,447). There was no significant difference between patch and patch+ofiyazis pattern for plasma level of MDA (p=0,119), erythrocyte activities of CAT (p=0,434), GSH-Px (p=0,375) and SOD (p=0,146).As a result, we found decreased erythrocyte GSH-Px and SOD activities in patients with AA, whereas plasma MDA level increased and erythrocyte CAT activity was the same as the control group. İncreasing level of MDA support the literature which indicates lipid peroxidation may have a role in the pathogenesis of AA. Increase in superoxide radicals may be responsible for decrease in SOD and GSH-Px activity.en_US
