dc.description.abstractAmaç: Meme kanseri geliĢmiĢ ve geliĢmekte olan ülkelerde oldukça sık görülen bir kanserdir. Meme kanseri tüm insanlarda, akciğer kanserinden sonra ikinci en çok görülen kanser olup, kadınların en sık kanseri ve kanserden ölüm nedenidir. Meme kanserinin oluĢumunda rol oynayan genetik faktörlerden bir tanesi de genetik polimorfizmlerdir. Bu çalıĢmada meme kanserli hastalar ve normal populasyonda hTERT geni rs2736100 bölgesindeki polimorfizim sıklığını ve bu durumun meme kanserli hastalarda kliniopatolojik bulgularla iliĢkisini araĢtırdık.Yöntem: ÇalıĢmaya Mustafa Kemal Universitesi Tıp Fakültesi‟nde , takip ve tedavi edilen 86 meme kanserli hasta ve bunların yaĢ uyumlu sağlıklı kontrolleri dahil edildi. Hastalardan ve yaĢ uyumlu sağlıklı kontrollerinden 2 cc kan alındı ve pcr ile analizi ile rs2736100 gen polimorfizmi bakıldı.Bulgular: Hastaların yaĢ ortalaması 52,7 idi. ÇalıĢmaya katılan 86 meme kanser hastalarının %76,3‟ünde metastaz yokken, %23,7‟sinde metastaz mevcuttu. Meme kanseri grubunda rs2736100‟deki nükleotid dizilimleri; AC %60,5, AA%26,7, CC %12,8, kontrol grubunda AA %47,7, AC%32,6, CC %19,8, olarak bulundu. Ġki grup arasında genotip dizilimleri açısından istatistiksel olarak anlamlı fark vardı (p=0,001). Meme kanserli hastarın hTERT genindeki rs2736100 polimorfizmi ile kanser tipi, kanserin evresi, lenf nodu tutulumu, metastaz varlığı, metastaz bölgeleri, kanser grade, lenfovasküler invazyon, perinöral invazyon, östrojen ve progesteron reseptör durumu ve cerb2 arasında istatistiksel olarak anlamlı bir iliĢki bulunmadı (p>0,05).Sonuç: Meme kanserli hastalarda hTERT geni rs2736100 bölgesi normal populasyondan farklılık göstermekte ve bu durum meme kanseri geliĢiminde rol oynayan faktörlerden birisi olabileceğini düĢündürmektedir.Anahtar kelimeler: Meme kanseri, rs2736100, hTERT
dc.description.abstractBackround and Aim: Breast cancer is seen most frequently in developed or developing countries. Breast cancer is most common cancer in humans following lung cancer and the most common cancer in women resulting mortality from cancer. Genetical polymorphisms are one of the genetical factors which plays apart in the development of the breast cancer. In this study we have investigated the polymorphism frequency of the hTERT gene rs2736100 regionamong normal and breast cancer diagnosied patients and its relation between both clinic aland pathological findings in breast cancer diagnosed patients.Methods: 86 breast cancer diagnosed patients who are treated and observed in Mustafa Kemal University school of medicine and their age equivalent control groups are involved in this study. Two cc blood samples are obtained both from patients and age equivalent healhty control group and by using pcr rs2736100 polymorphism was investigated.Results: The mean vaule of the patients was 52.7. Among the 86 patients who are involved in this study 76.3% of them have no metastasis and 23.7% of them had. In the breast cancer group nucleotide squence in rs2736100 region was found as ; AC %60.5, AA%26.7, CC %12.8 and in the control group AA %47.7, AC %32.6, CC %19.8 respectively. Between two groups there was a statisticaly significant difference in genotype squences (p=0,001). No statisticaly significant data was found in breast cancer patients with rs2736100 polymorphism in hTERT gene between type of the cancer, stage of the cancer, lymph node involment, presence of metastasis, metastasis regions, grade of the cancer, lymphovascular invasion, perineural invasion, oestrogen and progesterone receptor status and cerb-2 (p>0.05).Conclusion: HTERT geners 2736100 region in breast cancer patients is diffrent from the healhty ones and this situation can be considered as this polymorphism could be one of the factors in the development of the breast cancer.en_US
