dc.description.abstractVI-ÖZET Yıkanmış insan trombositlerinde protein ve glikoproteinler 3AVİS poli- akrilamid gel elktroforezi (PAGE), sodyum dodesil sülfat poliakrilamid gel elektroforezi (SDS-PAGE) yöntemleriyle incelendi PAGE gellerindeki proteinler Comassie parlak mavisi, glikoproteinler ise periodik asid ^'chiff ile boyanda» DAVIS-PAGE yönteminde gellerde.7 protein bandı görüldü. Adrenalinin proteinlerin konsantrasyonlarını etkilediği ve bromokriptinin hem adrenalin ile trombosit agregasyonuna mani olduğu hem de trombosit protein konsantrasyo nunu kontrol değerlerine doğru kaydırarak adrenalin etkisini nötralize ettiği görüldü. SDS-PAGE yöntemiyle 1:157,000, 11:132,000, 111:123,000, IV:120,000, V:1İ2,000, VI: 106,000, VII.66.000, VIII: 46,000, IX:4 3,000 X: 3 4,000 moleküler ağırlıkta. protein bandı elde edilirken, SDS-PAGE gellerinde ancak 5 glikoproxein bandı tesbit edildi. Bunların 1:132,000, 11:125,000, 111:1.12,000, IV:66,O00, V:52,000, molekül ağırlıklarında olduk ları hesaplandı ve GPIc.GPIIb, GPIIIa,GPIIIb ve GPVII olarak düşünüldü.

Glycoproteins by Polyacrylamide Gel Electrophoresis Kethods The washed human whole platelet proteins and glycoproteins were analyzed by DAVIS polyacrylamide gel electrophoresis (PAGE) and;sodium dodesyl sulphate > polyacrylamide gel electrophoresis (SDS-PAGE) methods. The proteins on the PAGE gels were dyed by Comassie brillian blue and glycoproteins by periodic acid gchiff reagent. 7 protein bands were detected on DAVIS-PAGE gels from platelets isola ted from normal donors Adrenalin effected. Adrenalin effected the concentra tion of these proteins and bromokriptin which inhibited the adrenalin induced platelet aggregation also reversed the effect of adrenalin on platelet protein concentration close to the control levels, neutralizing the effect of adrenalin- IP protein bands were detected on SDS-PAGE gels of platelets isolated from normal donorg(n:6) with molecular weights corresponding to I: 157,000, 11:132,000, 111:123,000, IV:120,000, Vill2,000, VI:l06.pC0r 711:66,000, VII 1:46,000, IX:43,000, X:34,0O0 5 glycoprotein bands were detected on SDS-PAGE gels of platelets isolated from normal donors (n:£) with molecular weights corresponding to 1:132,000, 11:125,000, 111:112,000, IV:66,000» V:52,000, and they were thought to be platelet GPIc, GPIIb, GPIIIatGPIIIb and GPVII respectively.en_US
