dc.description.abstractİmplantasyonda birçok sinyal molekülünün, blastosist ile reseptiv endometrium arasındaki haberleşmede rol aldığı bilinmektedir. Bu çalışmada, Wnt sinyal yolağı proteinlerinin östrus ve gebelik endometriumundaki ekspresyonları, immünfloresan ve real time PZR yöntemleriyle gösterilerek endometriumda oluşan değişikliklerdeki rolleri ve çeşitli sinyallerle nasıl düzenlendiği belirlenmiştir. Diöstrus evresinde uterus epitelinde Wnt-3 ve beta-katenin, endometrial bazal stromada Wnt-3, Wnt-7a, kaspaz-9 ve TCF, kan damarlarında ise Wnt-7a, kaspaz-9 ve TCF molekülleri kuvvetli ekspresyon göstermiştir. LEF-1, en düşük ekspresyona sahiptir. Metaöstrus evresinde Wnt-7a, uterus epiteli hücreleri ve diğer alanlarda da çok kuvvetli lokalize olmuştur. Östrusta, LEF-1 ve TCF, perimetrial-miyometrial bağ dokuda lokalizasyon göstermiştir. Uterus epitelinde, beta-katenin ve kaspaz-9, bez epitelinde beta-katenin, subluminal stromada, Wnt-3 endometrial bazal stromada, Wnt-3 Wnt-7a ve kaspaz-9, kan damarlarında ise Wnt-3, Wnt-7a ve kaspaz-9 çok kuvvetli eksprese olmuştur. Gebeliğin 7. gününde LEF-1 yalnız mezometrial desiduada, TCF ise yalnız anti-mezometrial desiduada lokalizasyon göstermiştir. Gebeliğin 8. gününde, primer desidualizasyon zonunda Wnt-3, Wnt-7a, beta-katenin ve kaspaz-9 ekpresyonu maksimumdur. Gebeliğin 9. gününde, tüm moleküllerin lokalizasyonları, hem anti-mezometrial desiduada hem de mezometrial desiduada belirgin düzeyde artış göstermiştir. Real time PZR sonuçlarına göre Wnt-3 en iyi ekspresyonu gebeliğin 9. gününde gösterirken, en düşük östrustta görülmüştür. Wnt-7a, gebeliğin 7. 8. ve 9. günlerinde az çalışmıştır. Beta-katenin tüm bu moleküller arasında en yüksek ifadeye sahip olup siklusta ve gebelikte en iyi çalışan proteindir. Kaspaz-9 ise, en iyi östrus ve metaöstrus dönemlerinde eksprese olmuştur. Sonuç olarak, peri-implantasyon süreci boyunca Wnt moleküllerinin (Wnt-3, Wnt-7a, beta-katenin, kaspaz-9, Tcf, Lef-1) sıçan endometriumunda hücre sinyalizasyonu, transkripsiyon ve hücre ölümünün belirlenmesinde önemli bir rolü olduğu saptanmıştır.Anahtar Kelimeler: Rat, Wnt Sinyal Yolağı Proteinleri, İmplantasyon, Östrus veGebelik Endometriumu, İmmünflorasan, Real Time PZR
dc.description.abstractIt is well known that plenty of signal molecules play important roles between the receptive endometrium and the blastocyst during implantation. In the present study, expressions of Wnt signal pathway proteins were demonstrated in the oestrus cycle and in the pregnancy endometrium by immunflourescence and real time PCR techniques therefore their possible roles and interactions with several other signals on endometrial alterations have been determined. It has been shown in the dioestrus cycle that Wnt-3 and β-catenin in the uterine epithelium, Wnt-3, Wnt-7a, caspase-9 and TCF in the basal stroma and Wnt-7a, caspase-9 and TCF in the blood vessels were all expressed strongly whereas LEF-1 had the lowest immunlocalization in those areas. Wnt-7a was expressed very strongly in uterine epithelial cells and in other areas during the metaoestrus cycle. LEF-1 and TCF were localized only in the perimetrial-myometrial connective tissue during the oestrus period. The strongly immunlocalized molecules and their localization areas during the oestrus period were as follows; β-catenin and caspase-9 in the uterine surface epithelium, β-catenin in the glandular epithelium, Wnt-3 in the subluminal stroma, Wnt-3, Wnt-7a and caspase-9 in the endometrial basal stroma, Wnt-3, Wnt-7a and caspase-9 in the blood vessels. While LEF-1 was expressed only in the mesometrial decidua, TCF was expressed only in the anti-mesometrial decidua during the seventh day of pregnancy. Highest Wnt-3, Wnt-7a, β-catenin and caspase-9 expressions were determined in the primary decidualization zone on day 8 of pregnancy. An increased immunolocalizations of all related molecules were determined both in the anti-mesometrial decidua and in the mesometrial decidua on day 9 of pregnancy. According to real time PCR findings, while Wnt-3 demonstrated its' highest expression on day 9 of pregnancy, it was found to be lowest in the oestrus. Wnt-7a had weak expressions on days 7, 8, and 9 of pregnancy. On the other hand β-catenin had the highest expression levels among all studied molecules thus it was the only molecule that worked both during the oestrus cycle and pregnancy. Caspase-9 was highly expressed during the oestrus and metaoestrus periods. In conclusion, it could be postulated that Wnt molecules might play crucial roles in cell signalling, transcription and cell death in the rat endometrium throughout the peri-implantation process.Key Words: Rat, Wnt Signal Pathway Proteins, Implantation, Oestrus and PregnancyEndometrium, Immunoflourescence, Real Time PCRen_US
