dc.description.abstractUzun kemiklerdeki segmental kemik defektlerinin tedavisi gunumiizde de sorun olarak kar§imiza 9ikmaktadir.(^ali§mamizda PLP/bBMP bilesjminin si9an radiuslannda olu§turulan genis, segmental defektlerin onanlmasinda sagladigi iyile§me degerlendirilmi§tir. Bu 9ah§mada 24 wistar cinsi yeti§kin erkek sKjan radiuslannda,bilateral olarak radius uzunlugunun %50'si oraninda osteoperiosteal segmental defekt olu§turuldu ve fareler A,B ve C olarak 3 gruba aynldilar. A grubundaki si9anlann sag radius defektlerine yiiksek doz bBMP i9eren PLP/bBMP kompoziti yerlesjtirildi, sol taraftaki defekt ise kontrol amaciyla BSA ile greftlendi. B grubundaki si9anlann sag taraftaki defektleri salt bBMP ve sol taraftaki defektleri ise salt PLP ile greftlendi. C grubunda yer alan si9anlann sag radiustaki defektleri dii§uk doz bBMP i9eren PLP/bBMP kompoziti ile greftlendi. Postoperatif 7.haftadabiitiin si9anlar oldiiruldu.Defektlerdeki kemik olu§umu radyolojik ve histolojik yontemlerle incelendi. Dii§iik yada yiiksek dozda olsun bBMP i9eren PLP/bBMP kompoziti ile greftlenen defektlerde kemik rejenerasyonu goriildii. BSP veya salt PLP yada salt bBMP i9eren greftlerin yerle§tirildigi defektlerde kemik olu§umu gozlenmedi. Test ve kontrol gruplan arasmda istatistiksel olarak anlamli fark bulundu (p <0.05).I§ik mikroskobu ile degerlendirmede inflamasyonun histolojik bulgulanna rastlanilmadi.Bu 9ali§mamizm sonu9lan ile PLP/bBMP kompozitinin klinik olarak lokal kemik olu§umuna gereksinim duyuldugu durumlarda kemik greftine kar§i iyi bir se9enek ve biiyuk bir potansiyel oldugu desteklenmektedir. dc.description.abstractThe repair of segmental bone defects remains a dilemma even to the present day. In this study the results of a composite of PLP/bBMP on the healing of large segmental radial defects in rats was considered. 24 adult male wistar rats were divided evenly into three groups A through C and an osteoperiosteal defect 50 % of the length of the bone was made bilaterally in each rat. In group A, the right radial defect was grafted with a high dose bBMP composite of PLP/bBMP, the left radial defect was grafted with BS A which served as control. In group B the right radius was grafted with bBMP whilst the left radius was grafted with PLP alone. In group C only the right radius was grafted with a low dose bBMP PLP/bBMP composite. All rats were killed after 7 weeks postoperatively.The formation of bone in the defects was evaluated by radiological and histological methods. Defects that had been grafted with either high or low PLP/bBMP composite showed regeneration of bone Defects that had been grafted with BSA, PLPor bBMP alone showed no formation of bone.There was found to be a statistically significant difference between the test and control groups. No inflammation was seen with the light microscope and only traces of PLP could be detected in defects that had been grafted with the composite.The results of this study support the possible application of bBMP/PLP composite as a bone graft substitute which can be of great potential for clinical states neccessitating local bone formation.Key Words: PLP, bBMP, Bone defecten_US
