dc.description.abstractIn this study, we investigated the effects of portal hypertension on gastric motor and secretoryfunctions in an experimental model. At the same time, we also searched if there was any relationbetween the gastric funtion and polymorphonuclear cell infiltration to the gastric wall. Moreover,we investigated the role of endothelin in all of these pathophysiologic events. The study wasaccompolished on Wistar-Albino female rats weighing 150-250 g. Animals were divided into fivegroups. Control, sham operated, ET-A receptor blocker-treated, portal hypertensive, portalhypertensive+ET-A receptor blocker treated rats were subjected to tests of gastric secretory,motor, mucosal function and gastric wall myeloperoxidase (MPO) activity was measured as anindex of polymorphonuclear infiltration. Portal hypertension (PHT) was induced by the partialportal vein ligation (>10 mmHg). BQ-485, a specific ET-A receptor antagonist was given of1J.lglkg I.V. for three days. Portal hypertension suppressed the gastric acid and total fluidsecretion and decreased the gastric emptying. We also observed an increase in mucosalpermeability but no alteration was evident in gastric wall MPO activity. The effects ofPHT ongastric secretory, motor and mucosal functions were reversed by the treatment of ET-A receptorblocker.We conclude that PHT suppresses the gastric motor and sectretory functions and endothelin playsa role in the pathophysiology of the PHT.en_US
