dc.description.abstractBu çalışmada, Lakkaz enzimi (EC 1.10.3.2), poli(vinil alkol)-kalsiyum aljinat (PVA-CaAlj), poli(N-izopropilakrilamit)-kalsiyum aljinat (P(NIPA)-CaAlj) kürelerine, karbodiimit (CDI), N-hidroksisüksinimit (NHS), karbodiimit ve N-hidroksisüksinimit (CDI-NHS) ile aktifleştirilerek kovalent bağlanma yöntemiyle immobilize edildi. Serbest enzim için Michaelis-Menten sabiti (Km) ve maksimum tepkime hızı (Vmak) değerleri sırası ile 1,70x10-2mM, 2,08 x10-3 mM.dak-1 olarak bulundu. İmmobilize enzimler için ise Km değerleri 2,87x10-2 mM ? 5,55x10-2 mM arasında değişirken Vmak değerleri 5,30x10-3 mM.dak-1 - 8,58x10-3 mM.dak-1 aralığında bulundu. Serbest enzim ve immobilize enzimler için optimum pH değerleri sırasıyla 5,0 ve 6,0 ve optimum sıcaklık değerleri sırasıyla 40ºC ve 45ºC olarak bulundu. Serbest enzim 34 gün sonunda aktifliğinin %60'ını korurken immobilize enzimlerinin ise 30-32 gün sonunda aktifliklerinin yaklaşık %80-87'sini koruduğu görüldü. İmmobilize enzimlerin 5 kez kullanım sonunda ve enzim aktifliklerini %86-92 aralığında koruduğu bulundu.
dc.description.abstractIn this study, Laccase (E.C 1.10.3.2) was covalently immobilized onto poly(vinylalcohol)-calcium alginate (PVA-CaAlj), poly(N-isopropylacryl amide) -calcium alginate (P(NIPA)-CaAlj) beads by activating with carbodiimide (CDI), N-hydroxysuccinimide (NHS), carbodiimide and N-hydroxysuccinimide (CDI-NHS). Michaelis-Menten constant (Km) and maximum reaction rate (Vmax) values were found as 1.70x10-2mM and 2.08x10-3mM.min-1 for free enzyme, respectively. Km and Vmax values were changed between 2.88x10-2 mM ? 5.56x10-2 mM and 5.30x10-3 mM.min-1 ? 8.58x10-3 mM.min-1 for immobilized enzymes, respectively. Optimum pH and temperature were observed as 5.0, 6.0 and 40oC, 45oC for free enzyme and immobilized enzymes, respectively. After 34 days of storage at 4oC free enzyme retained 60% of its original activity and immobilized enzymes were retained 80-87% of their original activities, after 30-32 days of storage at 4oC. İmmobilized enzymes were used repeatedly 5 times, were retained 86-92% of their original activities.en_US
