dc.description.abstractBu araştırma Ağustos 2009 ve Eylül 2010 tarihleri arasında Kırıkkale ilinden alınan bir orman faresi, Apodemus sylvaticus örneğinin sitokrom b ve ß-aktin genlerinin belirlenmesine dayanmaktadır. Kulak kepçesinden alınan doku örneğinin DNA izolasyonu yapılmış ve mitokondriyal sitokrom b ile ß-aktin genleri Türkiye'de ilk defa Polimeraz Zincir Reaksiyonu ile çoğaltılarak, sırasıyla 139 ve 154 baz çifti uzunluğunda dizin elde edilmiştir. Sitokrom b genine ait bazlardan Adenin % 25,89, Timin % 31,65, Guanin % 13,66 ve Sitozin % 28,05 olarak; ß-aktin genine ait bazlardan Adenin % 22,07, Timin % 21,42, Guanin % 25,32 ve Sitozin % 31,16 olarak tespit edilmiştir. Sonuçlar GenBankası verileriyle karşılaştırılmış ve sitokrom b geni dizini Apodemus'un türleriyle % 96 oranında benzerlik gösterdiği; ß-aktin gen dizini ise Rattus norvegicus ile % 96 oranında benzerlik gösterdiği tespit edilmiştir.Anahtar Kelimeler: Orman Faresi, Apodemus sylvaticus, Sitokrom b geni, ß-aktin geni, Türkiye.
dc.description.abstractThis study is based on determination of cytochrome b and ß-actin genes of forest mouse, Apodemus sylvaticus from Kırıkkale Province between August 2009 and September 2010. The DNA isolation of tissue sample which was obtained from auricle was carried out and mitochondrial cytochrome b and ß-actin genes was amplified via Polymerase Chain Reaction fort he first time in Turkey and 139 and 154 base pair were obtained respectively. Adenine 25,89 %, thymine 31,65 %, guanine 13,66 % and cytosine 28,05 % belongin to cytochrome b gene was determined and adenine 22,07 %, thymine 21,42 %, guanine 25,32 %, cytosine 31,16 % ß-actin gene. Results were compared with that of Gene Bank and cytochrome b gene was resembles Apodemus species with 96 % ratio. But ß-actin gene was also resemble Rattus norveqicus with 96 % ratio. Diagnostic characters and some habitat features were recorded.Key words: Wood mouse, Apodemus sylvaticus, cytochrome b gene, ß-actin gene, Turkey.en_US
