Papaver dubium L. subs. laevigatum (M.Bieb.) Kadareit türünün alkoloitleri üzerinde araştırmalar

Abstract

V. ÖZET- 43 - Bu çalışmada Bolu-Gerede'den toplanan Papaver dubium subsp. laeavigatum (M. Bieb) Kadereit türü alkaloitleri yönünden ilk kez incelenmiştir. Bitkinin toprak üstü kısımlarından elde edilen tersiyer alkaloit ekstresi üzerinde çalışılmış, bu alkaloitler sütun kromatografisi ve preparatif ince tabaka kromatografisi ile birbirinden ayrılmış ve saflaştırılmıştır. Bu türde majör alkaloit olarak protoberberin grubundan katerner yapıdaki berberine, minör alkaloit olarak yine protoberberin grubundan katerner yapıdaki thalifendine, aporfin grubundan isocorydine ve corydine izole edilmiştir. Protoberberin grubundan katerner yapıdaki thalifendine Papaveraceae familyası için yeni bir alkaloittir. Elde edilen alkaloitlerin yapılan ince tabaka kromatografisindeki Rf değerlerinin şahit maddeler ile karşılaştırılması ve spektral değerler (UV, İR, !H-NMR ve EIMS) ve literatür verileri ile tayin edilmiştir. Sonuç olarak Papaver lacerum türü ile sinonim kabul edilen Papaver dubium subsp. laevigatum türünün bu çalışmada alkaloitleri bakımından farklı olduğu saptanmıştır.

VI. SUMMARY- 44 - In this study, the alkaloids of Papaver dubium subsp. laevigatum (M. Bieb) Kadereit which was collected from Bolu-Gerede were investigated for the first time. Total tertiary alkaloidal extract was prepared from the aerial parts of the plant sample and the alkaloids were isolated and purified by column and preparative thin layer chromatographies. A quaternary protoberberinium salt berberine was isolated as a major alkaloit. The other quaternary protoberberinium alkaloit thalifendine has been isolated as a minor alkaloit together with isocorydine and corydine which belong to aporphine group. The presence of thalifendine has been shown in Papaveraceae family for the first time. The structure of alkaloids was confirmed by comparing their Rf values on thin layer chromatography with authentic samples, spectral datas (UV, ^-NMR, EIMS, FAB) and literature findings. In conclusion, it has been shown that alkaloids of Papaver dubium subsp. laevigatum which is considered as synonym of Papaver lacerum are different from those of Papaver lacerum.
