dc.description.abstractAmaç: Çalışmamızın amacı farklı HPV alt tiplerinin yapmış olduğu E6 ve E7 kanser yapıcı protein tespiti ve servikal displaziler üzerine etkilerinin araştırılmasıdır.Materyal-Metod: : Smear sonuçları ASCUS, ASC-H, LSIL, HSIL ve AGC olarak raporlanan 21-65 yaşları arası 77 hasta dahil edilmiştir. HPV DNA'nın tespit ve tiplendirilmesi HPV sign® Q24 complete kiti, RotorGene ve PyroMark Q24 (Qiagen, Almanya) sistemleri ile kullanılarak yapıldı. HPV DNA pozitif olguların servikal biyopsilerinden hazırlanan örneklerde HPV E6/E7 mRNA gen ekspresyonu NASBA yöntemiyle, NucliSENS® EasyQ Genetic Analyzer cihazı (Biomerieux, Fransa) kullanılarak araştırıldı.Tartışma: HPV DNA pozitif olguların servikal biyopsi sonucunda %45,5 (10/22)'inde anormal servikal lezyon (CIN 1,2,3) tespit edilirken, %13,63'ünde CİN 3 servikal lezyon görülmüştür. HPV mRNA pozitif olguların servikal biyopsi sonucunda %45,5 (5/11)'inde anormal servikal lezyon (CIN 1,2,3) tespit edilirken, %27'sinde CİN 3 görülmüştür. HPV DNA ve HPV mRNA markerlarının ikiside pozitif olan grupların servikal biyopsi sonuçları CİN3 olanları karşılaştırıldığında istatistiksel olarak anlamlı farklılık bulunamamışıtr (p=0,375). Ama HPV mRNA pozitif olan gruptaki CİN 3 oranı yüksekliği dikkat çekmektedir.Sonuç: Servikal dokuda HPV E6/E7 mRNA saptanmasında kullanılan testler, servikal displazi şiddetiyle daha iyi korelasyon gösterir. Bu testler serviks kanserini öngörmede önemli bir potansiyele sahiptir. Fakat konu üzerinde daha geniş ölçekli çalışılmaların yapılmasına ihtiyaç vardır. dc.description.abstractObjective: The objective of our study is to determine E6 and E7 carcinogenic proteins caused by different sub-types of HPV as well as investigating their effects on cervical dysplacias. Material-Method: 77 patients aged between 21-65, whose Smear results were reported as ASCUS, ASC-H, LSIL, HSIL and AGC, were incorporated into the study. The identification and typology of HPV DNA were performed by using HPV sign® Q24 complete kit and RotorGene and PyroMark Q24 (Qiagen, Germany) systems. HPV E6/E7 mRNA gene expression within the samples prepared from the cervical biopsies of HPV DNA positive cases was investigated through NASBA method (Nucleic acid sequence based amplification) by using NucliSENS® EasyQ Genetic Analyzer device (Biomerieux, France).Discussion: As the result of the cervical biopsies of HPV DNA positive cases, an abnormal cervical lesion (CIN 1,2,3) was detected in 45,5% (10/22) of these cases, whereas a CIN 3- cervical lesion was seen in 13,63% of them. As the result of the cervical biopsy results of HPV mRNA positive cases, an abnormal cervical lesion (CIN 1,2,3) was detected in 45,5 % (5/11) of these cases, while in 27% of them was CIN 3 seen. When the cervical biopsy results of the groups, the HPV DNA and HPV mRNA markers of which were both positive, were compared with those having CIN 3, no statistically significant difference could be found (p=0,375). However, the high rate of CIN 3 within the group that is HPV mRNA-positive attracts attention. Result: The tests used for determining HPV E6/E7 mRNA in the cervical lesion show a better correlation with the severity of cervical dysplasia. These tests have a significant potential in foreseeing cervical cancer. Yet, studies on a larger scale are required to be performed on this subject.
