Adana atmosferinden izole edilen küf mantarlarından alerjen üretimi

Abstract

Bu çalışmada, bölgesel (Adana atmosferi izolatı) küflerden deri delme (skin prick) testinde kullanılmak amacıyla test materyali üretilmiş ve standart alerjenler ile kıyaslanmıştır. Cladosporium cladosporioides, Cladosporium herbarum ve Alternaria alternata olarak tanılanmış örnekler PDA katı besiyerinde tek koloni düşecek şekilde, saf kültürler ise Czapek Dox sıvı besiyerinde 26°C'de 14 gün üretilmiştir. Elde edilen miseller filtre kağıdı ile toplanarak 1 gece kurutulup sıvı azot, ultra turrax, sonikatör ve dondur-çöz uygulamalarına tabi tutulmuştur. Son olarak liyofilizatörde 24 saat liyofilize edilmiştir. Ekstraktların protein miktarı Bradford yöntemi ile tayin edilmiştir. Her suşa ait ekstraktlar kolon kromatografisi ile fraksiyonlarına ayrılmıştır. Total ekstraktlar ve fraksiyonların protein bantları ile moleküler ağırlıkları %10'luk SDS-PAGE analizi ile saptanmıştır.Üretilen test materyalleri Çukurova Üniversitesi Tıp Fakültesi Çocuk İmmünoloji ve Alerji Hastalıkları Bilim Dalı Kliniği'nde 37 hastaya uygulanmıştır. Cladosporium cladosporioides için 5 hastada, Cladosporium herbarum için 9 hastada, Alternaria alternata için 35 hastada hem standart hem de üretilen test materyali ile test sonucu pozitif sonuçlar alınmıştır. Bulgulara göre üretilen test materyalinin klinikte teşhis amaçlı kullanılabileceği önerilebilir.

In this study, test material was produced of regional molds for use in skin prick test and compared with standard allergens. Samples identified as Cladosporium cladosporioides, Cladosporium herbarum and Alternaria alternata were cultured on PDA solid medium for the production of single colonies and the pure colonies were produced on Czapek Dox liquid medium at 26°C for 14 days. The obtained mycelium was collected with filter paper and dried for 24 h and subjected to liquid nitrogen, ultra turrax, sonicator, freeze-thaw applications. Finally, the extracts were lyophilized for 24 h. The protein content of the extracts was determined by the Bradford Method. Extracts of each strain was fractionated by Column Chromatography. Protein bands and molecular weights of total extracts and fractions were determined by 10% SDS-PAGE analysis.The test materials produced were applied to 37 patients at the Department of Pediatric Immunology and Allergic Dieases Clinic of Çukurova University Medicine Faculty. In 5 patients for Cladosporium cladosporioides, test results were positive with both standard and produced test material, 9 patients for Cladosporium herbarum; positive with standard and test material, for Alternaria alternata in 35 patients; positive with standard and test material. It can be suggested that the test material produced according to the findings may be used clinically for diagnostic purposes.
