Abstract
Bu çalışmada sıçanlarda kurşun asetat ile oluşturulan oksidatif strese karşı geraniolün koruyucu etkisini araştırmak amaçlandı. Çalışma için, kontrol grubu (K), kurşun asetat (KA) grubu, geraniol (G) grubu ve kurşun asetat + geraniol (KA+G) grubu olmak üzere dört farklı grup oluşturuldu. Sıçanların karaciğerinde MDA, redükte GSH ve çeşitli minerallerin düzeyleri ile AChE, GST, CAE aktiviteleri belirlendi. Elde edilen sonuçlara göre KA uygulama grubunda malondialdehit (MDA) düzeyi K grubuna göre önemli düzeyde artarken (p<0.001), geraniolün etkisiyle KA+G grubunda MDA düzeyi KA grubuna göre önemli düzeyde azaldı (p<0.01). KA grubunda redükte GSH düzeyinde K grubuna göre nisbi bir azalma gözlenmesine rağmen, bütün uygulama gruplarının redükte GSH düzeylerinde K grubuna göre istatistiksel olarak önemli bir fark gözlenmedi. AChE, GST ve CAE enzim aktivitelerinin ise KA grubunda, K grubuna göre önemli düzeyde azaldığı belirlendi (p<0.01; p<0.001). Ayrıca, kurşun düzeyinin KA grubunda K grubuna göre önemli düzeyde arttığı, KA+G grubunda ise KA grubuna göre önemli düzeyde azaldığı belirlendi (p<0.001). Sonuç olarak, kurşun asetat ile oluşturulan oksidatif strese karşı geraniol'ün antioksidan olarak olumlu bir etki gösterdiği ve seçilen biyokimyasal belirteçlerin de bu tür çalışmalar için kullanışlı olduğu sonucuna varılmıştır. In this study, it was aimed to investigate the protective effect of geraniol against oxidative stress produced by lead acetate in rats. For the study, four different groups as control group (C), lead acetate (LA) group, geraniol (G) group and lead acetate + geraniol (LA + G) group were formed. MDA, reduced GSH and various minerals levels and AChE, GST, CAE activities were determined in the liver of rats. According to the results, MDA level was significantly increased in LA group compared with C group (p<0.001) whereas MDA level was significantly decreased in LA+G group compared with LA group due to effect of geraniol (p<0.01). There was no statistically significant difference in the reduced-GSH levels of all treatment groups compared to the C group, although a relative decrease in the GSH level of the LA group compared to the C group was observed. AChE, GST and CAE enzyme activities were significantly decreased in the LA group compared to the C group (p<0.01; p<0.001). It was also determined that the lead level was significantly increased in the LA group compared to the C group and significantly decreased in the LA + G group compared to the LA group (p<0.001). As a result, it has been concluded that geraniol as an antioxidant against oxidative stress induced by lead acetate has a positive effect and selected biochemical markers are useful for such studies.
