Abstract
ÖZET Yüksek Lisans Tezi ÇİFT FAZLI SİSTEMDE 2-FENİL ETANOLUN FENİL ASET ALDEHİTE BİYOTRANSFORMASYONU Devrim ÇELİK Ankara Üniversitesi Fen Bilimleri Enstitüsü Kimya Mühendisliği Anabiüm Daü Danışman : Prof. Dr. Ülkü MEHMETOĞLU Aromatik aldehitlerden biri olan fenilasetaldehit gıda ve kozmetik sanayilerinde kullanılan hoş kokulu bir tatlandırıcıdır. Kimyasal üretiminin yerine, biyoteknolojik yöntemlerden biri olan biyotransformasyon ile üretimi yapılmaktadır. Biyotransformasyon; biyokatalizör ile A substratınm B ürününe dönüştüğü bir reaksiyondur. Çift fazlı sistemde gerçekleştirilen biyotransformasyonda, substrat ve ürünün seçimli olarak çözünebildiği organik bir çözücünün kullanımı, ürün veya substrat inhibisyonunu azaltır ve ürünü organik âzda geri kazanma olanağı sağlar. Çift fazlı sistemde 2-feniletanoIden fenilasetaldehit üretiminin amaçlandığı bu çalışma, sıcaklık ve karıştırma hızı kontrollü orbital çalkalayıcıda, 150 mL lik erlenlerde gerçekleştirilmiştir. 2-feniletanol ve fenilasetaldehit derişim leri gaz krom otografi cihazı ile analizlenm iştir. Çalışmanın ilk aşamasında, çözücü olarak izo-oktan seçilmiştir. Substrat suda çözünürken ürün olan fenilase&ldebitin çözücü fazına alınarak uzaklaştırılması ve böylelikle üretim veriminin arttırılması amaçlanmıştır. 2-feniletanol ve fenilasetaldehitin dağılma katsayısı üzerine değişik parametrelerin etkisi incelenmiş, süre ve karıştırma hızının etkisinin olmadığı, derişim ve sıcaklığın artmasıyla dağılma katsayısının azaldığı görülmüştür. Çalışmanın ikinci aşamasında, 2-feniletanolün fenilasetaldehite tek ve çift fazlı sistemde bryotransformasyonunda başlangıç substrat derişimi ve biyotransformasyon süresinin etkileri incelenmiştir. Çift fazlı sistemde uygun âz oram(organik fazmhacmi/sulu fazın hacmi) değerinin 1olduğu bulunmuştur. Substrat inhibisyon kinetiği çıkarılmış ve kinetik sabitleri belirlenmiştir. Çalışmanın son aşamasında, substrat mhibisyonunu önlemek amacıyla kesikli ara beslemeli (fed-batch) işletim gerçekleştirilmiştir. Instead of chemical production, it is produced by biotransformation which is one of the biotechnological methods. Biotransformation, is the production of B product from A substrate by a bioeatafysL In two-phase biotransformation systems, me usage of an organic solvent in which the substrate or the product are preferentially solubilized, provides the opportunitiy to reduce the inhibition of the bioconversion and to recover the product in the organic phase. The study to produce phenlyacetaldeyde from 2- phenlyethanol in a two- phase system was performed in an orbital shaker and 150 m L shaken flasks. Concentrations of 2- phenlyethanol and phenlyacetaldeyde were determined by gas choromotography. In the first step of the study, iso- octane was choosen as the solvent It was aimed to solve the substrate in the aqueous phase, to remove phenlyacetaldeyde by taking it to the organic phase and to increase the product yield. The effect of various parameters on the partitioning coefficients of 2- phenlyethanol and phenlyacetaldeyde were invastigated and it was seen mat, time and agitation rate have no effect on partition coefficent. Partition coefficent decreased by increasing temperature and concentration. In the second step of the study, the effect of initial substrate concentrations and time on the biotransformaticHi of 2- phenlyethanol to phenlyacetaldeyde in aqueous and two- phase systems were investigated. The optimum phase iiiratio (organic phase volume/ aqueous phase volume) value of the two-phase system was found as I. Substrate inhibition kinetics were investigated and die kinetic constants were found. In the last step of me study, in order to prevent the substrate inhibition, fed- batch operation was done. The optimum conditions which effect the production of phenlyacetaldeyde by Response Surface Methadology (RSM) were found to be 3 fed-batch number, 2.75 g/L initial substrate concentration, 60 min time, ISO rpm agitation rate.
