Abstract
Kanser iyilestirme ve tedavisindeki son gelismeler, hücre sinyali iletilerinde rol alanbelirli tirozin kinazların inhibe edilmesi ve sonuc olarakta hücresel proliferasyon vetümör gelisiminin durdurulması üzerine odaklanmıstır. Sinyal transdüksiyoninhibitörleri kanser hücrelerinin gelisimi ve metastazı için gerekli olan sinyaltransdüksiyon olusumlarını inhibe etmek için kullanılır. Bu anti-kanser ajanlar;polimerik miseller, nanopartiküller, ve lipozomlar gibi kolloidal ilaç tasıyıcısistemlere yüklenebilmektedir ve daha sonra seçimli olarak, kanserli hücre yüzeyreseptörlerine karsılık gelen ligand veya antibadiler yardımı ile kanser hücrelerinedogrudan hedeflendirilebilmektedir. Çalısmanın ilk bölümünde gastrointestinalsistem kanserlerinde kullanılan bir C-Kit kinaz inhibitörü (WBZ-4) gelistirilmis veyüksek hidrofobisitesinden dolayı hücre içine tasınımı saglamak ve toksisiteyiönlemek amacıyla lipozomal bir tasıyıcı içine enkapsüle edilmistir. lacın aktivitesi;in vitro hücre proliferasyon çalısmaları, protein inhibisyon çalısmaları ve kinaztayinleri ile test edilmistir. Hücre farmakolojisi çalısmaları inhibitörün FITC ileisaretlenmesiyle in vitro olarak yapılmıstır. Çalısmanın ikinci bölümünde siRNAtasınımı için kullanılmak üzere iyonik jelasyon yöntemi ile kitosan nanopartiküllerhazırlanmıstır. Nanopartiküllerin fizikokimyasal ve morfolojik özellikleri; zetapotansiyel analizi, atomik kuvvet mikroskobu (AFM) ve taramali elektronmikroskobu (SEM) yöntemleri ile belirlenmistir. Konkanavalin A nanopartiküllerinseçimli olarak hücrelere yüklenmesini arttırmak amacı ile nanopartiküllerin üzerineimmobilize edilmistir. Kitosan nanopartiküllerin in vitro ve in vivo çalısmalarıgerçeklestirilmistir. Elde edilen sonuçlar WBZ-4-yüklü lipozomlar ile siRNA-yüklüve/veya konkanavalin A ile modifiye edilmis kitosan nanopartiküllerin kansertedavisi için güvenli, biyouyumlu ve verimli bir yol sagladıgını göstermistir.Anahtar Sözcükler: Kitosan nanopartikül, Konkanavalin A, Lipozom, Sinyaltransdüksiyon inhibitörü, siRNA, Hedefli kanser terapisi. Recent research in cancer therapy and treatment has focused on inhibiting certaintyrosine kinases involved in cell signaling pathways, thereby halting cellularproliferation and tumor growth. Signal transduction inhibitors are used to inhibit thesignal transduction pathways required for the growth and metastasis of cancercells. These anti-cancer agents can be associated with the colloidal drug carriersystems such as polymeric micelles, nanoparticles, and liposomes, which can thenbe actively targeted to specific tumor cells by means of ligands or antibodiesagainst tumor associated cell surface receptors. In the first part of the study aninhibitor of the C-Kit kinase (WBZ-4) in gastrointestinal stromal (GIST) tumors wasdeveloped and due to its high hyrophobicity encapsulated into liposomes topromote cellular delivery and prevent toxicity. The activity of the drug was testedby in vitro cell proliferation studies, protein inhibition studies and kinase assays.Cellular pharmacology studies were performed in vitro by labelling the compoundwith FITC. In the second part of the study, chitosan nanoparticles for siRNAdelivery were prepared by ionic gelation method. The physicochemical propertiesof the nanoparticles were determined by size, zeta potential analysis, atomic forcemicroscopy (AFM) and scanning electron microscopy (SEM). Concanavalin A wasimmobilized on chitosan nanoparticles to increase transfection efficiency and topromote targeting. In vitro and in vivo studies of chitosan nanoparticles wereperformed. Obtained results showed that liposomal-WBZ-4 and siRNA entrappedand/or concanavalin-A modified chitosan nanoparticles provide a safe,biocompatible and efficient means for the treatment of cancer.Keywords: Chitosan nanoparticle, Concanavalin A, Liposome, Signal transductioninhibitor, siRNA,Targeted cancer therapy.
