Abstract
Kontrollü ilaç salımı ilaç sektöründe büyüyen ve gelecek vadeden bir sektördür. İlacın hedeflenen noktaya direk ulaşması ve minimize edilmiş yan etkiler, kontrollü ilaç salımını geleneksel yöntemlere göre avantajlı kılmaktadır. Kil ile hazırlanan polimerik nonokompozitler, ilaç salımı çalışmalarında önemli bir yer tutmaktadır. Smektit kil grubuna ait olan montmorillonit (MMT) düzenli katmanlı yapısı sebebiyle polimerik nanokompozit hazırlanmasında sıklıkla tercih edilmektedir. Kitosan (CS) doğal biyodönüşümlü, birçok kil türüne yüksek ilgi gösteren bir polimerdir. Oldukça biyouyumlu olması, kabul edilebilir derecede biyodönüşümlü olması, düşük toksik özelliği bu kompoziti nanokompozit üretiminde cazip bir seçenek olmasına neden olmaktadır. Paroksetin hydrochloride (PHH) majör depresif bozukluklarda kullanılan bir antidepresandır. Bu çalışmada kitosan/montmorillonit kompozit filmleri çözelti döküm yöntemi ile hazırlanmıştır. Farklı kitosan/kil (MMT/NaMMT) (1/0, 1/0,1, 1/0,2, 1/0,4) oranlarında hazırlanan kompozitlere Paroksetin (30 mg) eklenmiştir. Ayrıca 1/0,4 oranında hazırlanan kitosan/kil (MMT/NaMMT) kompozitlere farklı miktarlarda Paroksetin (30 mg, 60 mg) ve gliserol (0, 25 mg, 50 mg) eklenmiştir. İlaçlı ve ilaçsız filmlerin yapısal özelliklerinin incelenmesi için FT-IR ve SEM analizleri ile şişme testleri gerçekleştirilmiştir. İlaç içeren bütün kompozitlerin ilaç salım profilleri 37 0C ve pH 7.4 olan fosfat tamponu içeren ortamda incelenmiştir. En yüksek ilaç salımı sadece kitosan/PHH biyokompozit ile 170 saatin sonunda %69 olarak gerçekleştirilmiştir. Kitosana eklenen kil oranı arttıkça salım miktarı ve hızı azalmıştır. Herbir kompozitten in vitro ilaç salım verisi dört farklı kinetik modele uyarlanarak (Korsmeyer_Peppas, Higuchi, Sıfırıncı derece, birinci derece), salım kinetiği parametreleri elde edilmiştir. Bu çalışmada hazırlanan kitosan/montmorillonit biyokompozitinin Paroksetin ilacı için uygun bir taşıyıcı olma potansiyeli taşıdığı gösterilmiştir. Controlled drug delivery is promising and growing segment of the pharmaceutical market. Targeted delivery and minimized side effect makes it advantageous than the traditional drug delivery method. Polymeric nanocomposites prepared with clay has great potential importance in drug release studies. Montmorillionette (MMT) which belongs to smectite clays, with a regular layered structure is widely used in preparing polymeric nanocomposites. Chitosan (CS) is natural biodegradable polymer and has extremely high affinity for many types of clays. Excellent biocompatibility and admirable biodegradability, low toxicity make it eligible for nanocomposites production. PHHetine hydrochloride (PHH) is a antideppresant, prescribed for major depressive disorders. In this study, chitosan/montmorillonite composite films were prepared by solvent casting method PHHetine (30 mg) was added to composites various chiton/clay ratio (MMT/NaMMT) (1/0, 1/0,1, 1/0,2, 1/0,4). In addition to that different amounts of PHHetine (30 mg, 60 mg) and glycerol (0, 25 mg, 50 mg) were supplemented to chitosan/clay composites prepared at 1/0.4 ratio. To investigate the structual properties of drug-containing and non-drug containing films FT-IR, and SEM analysis and swelling tests were done. The release profiles of all drug containing composites into phosfate buffer at 37 0C and pH 7.4 were investigated. The highest drug release was observed by CS/PHH nanocomposite as 69 % after 170 hours. Increasing in clay ratio incorporated into chitosan resulted by decreasing drug relase rate and amount. In vitro drug release data for each nanocomposites were fitted into four different kinetic models: Zero order, First order, Higuchi and Korsmeyer-Peppas models. In this study, it has been exhibited that chitosan/montmorillonit nanocomposite developed has a potential to become a suitable carrier for PHHetine drug.
