Abstract
Endometriozis, uterusun içini döşeyen ve endometrium olarak adlandırılan dokununuterus dışında bir yerde görülmesidir. Buralarda kronik inflamatuar reaksiyonuindükleyerek skar dokusu oluşmasına yol açar. Başlıca pelvik periton, overler,rektovajinal septum, mesane ve bağırsaklarda görülür. Nadiren de karaciğerde vediyaframda görülebilir.Endometriozis üreme çağındaki 10 kadından 1'nde görülür (çoğunlukla 15-49 yaşlarıarasında.Endometriozis bulunduğu yere, yaygınlığına ve endometriotik implantların derinliğine,adezyonların varlığına ve derecesine, ovaryan endometrioma bulunup bulunmamasınabağlı olarak Evre 1(minimal), Evre 2(hafif), Evre 3(orta), Evre 4(ağır) olarak 4 gruptasınıflandırılır.Bu çalışmada, hastalardan elde edilen endometriozis doku örnekleri spektroskopikyöntemlerden biri olan Raman Spektroskopisi ile incelenerek, endometriozisin hastadokuları üzerindeki moleküler düzeydeki etkileri saptandı. Deneysel olarak elde edilenDispersif Mikro Raman spektrumları hastalığın histolojik tanısından ve literatürden eldeedilen veriler ile karşılaştırılarak, hasta dokularındaki endometriozise bağlı değişiklikleryorumlandı.Bu çalışma, moleküler spektroskopisinin tıp alanında bir uygulamasıdır.Tez çalışmamız dahilinde incelediğimiz endometriozis hücre örneklerinin titreşimselspektral verilerine ulaşabilmek için, katı fazdaki endometriozis örneklerimizin dispersifRaman spektrumları oda sıcaklığında 532 nm ve 785 nm dalga boylu lazer kaynaklarınasahip olan bir Jasco NRS 3100 model mikro Raman spektrometre ile kayıt edildi.Kaydedilen spektrumlarda gözlenen karakteristik dikkat çekici bandlar işaretlendi.İşaretlenen bu bandların şiddet oranları 'Band Bileşenleri Analizi' yöntemi kullanılaraksaptadı. Bu oranlar yorumlanarak hasta dokuları arasındaki spektral farklılıklar(hastalığın şiddetine bağlı olarak ortaya çıkan) saptandı. Endometriosis is a condition in which tissue similar to the lining inside the uterus (called`the endometrium`), is found outside the uterus, where it induces a chronic inflammatoryreaction that may result in scar tissue. It is primarily found on the pelvic peritoneum, onthe ovaries, in the rectovaginal septum, on the bladder, and bowel. In very rare cases ithas been found on the diaphragm and in the lungs.Endometriosis affects an estimated 1 in 10 women during their reproductive years (lie.usually between the ages of 15 to 49).Endometriosis is classified into one of four stages (I-minimal, II-mild, III-moderate, andIV- severe ) depending on location, extent, and depth of endometriosis implants;presences and severity of adhesions; and presence and size of ovarian endometriomas.In thesis study, some tissue samples (with endometriosis) obtained from patients wereinvestigated by using one of spectroscopic methods Raman Spectroscopy, and therebythe effect of endometriosis (at molecular level) on the patient tissues were determined.Comparing the dispersive Raman spectra of the investigated samples, with the dataobtained from the histologic diagnosis and literature, the changes on the patient tissuesdue to endometriosis were interpreted.This work is a medical application of molecular spectroscopy.To obtain the vibrational spectral data of samples of endometriosis cells that weinvestigated in our thesis study, the dispersive Raman spectra of the endometriosissamples in solid phase were recorded at room temparature by using a Jasco NRS-3100model Micro-Raman spectrometer with laser sources of 532 nm and 785 nm wavelengths.The prominent ones among the characteristic fundemental bands observed in the recordedspectra were marked. The intensity ratios of these marked bands were determined byusing the method 'Band Component Analysis'. The spectral differences among theinvestigated patient tissues were determined by interpreting there ratios.
